不同时间和浓度大蒜素对卵巢癌细胞的影响及机制探究

摘要 目的探讨大蒜素(allicin)对人卵巢癌普通型HO-8910和高侵袭型HO-8910PM细胞株增殖的影响及诱导细胞凋亡机制、作用效果比较。方法不同浓度的大蒜素作用于人卵巢癌HO-8910和HO-8910PM细胞株不同时间,采用MTT法观察细胞活力;集落形成实验计算克隆形成率;PI/Hochest 33342荧光染色,检测caspase-3活性;Western blot法检测caspase-3和PARP蛋白表达。结果大蒜素能显著抑制HO-8910和HO-8910PM细胞的增殖和集落形成,影响caspase-3的活性和PARP蛋白裂解(P<0.05),呈时间和剂量依赖性;HO-8910PM细胞对大蒜素的敏感度均强于HO-8910细胞。结论大蒜素可显著抑制卵巢癌细胞增殖,可能通过调节caspase-3和PARP诱导细胞凋亡,且高侵袭性卵巢癌细胞更敏感。 Objective To investigate the effect of allicin on the proliferation of common type HO-8910 and highly invasive HO-8910PM cell lines of human ovarian cancer,and to compare the mechanism and effect of inducing apoptosis.Methods Human ovarian cancer cell lines HO-8910 and HO-8910PM were treated with different concentrations of allicin for different time.The cell viability was observed by MTT method.The colony formation rate was calculated by colony formation test.The activity of caspase-3 was detected by PI/Hochest 33342 fluorescence staining,and the expression of caspase-3 and PARP protein was detected by Western blot.Results Allicin could significantly inhibit the proliferation and colony formation of HO-8910 and HO-8910PM cells,affect the activity of caspase-3 and the cleavage of PARP protein in a time-and dose-dependent manner.The sensitivity of HO-8910PM cells to allicin was stronger than that of HO-8910 cells.Conclusion Allicin can significantly inhibit the proliferation of ovarian cancer cells,which may induce apoptosis by regulating caspase-3 and PARP,and highly invasive ovarian cancer cells are more sensitive.
