摘要
目的观察SNRPA1在胃癌中的表达情况及对胃癌细胞生长的影响。方法通过高通量差异基因增殖筛选技术(High Content Screening,HCS)分析发现,干扰SNRPA1基因可抑制胃癌细胞AGS增殖。利用TCGA数据库分析SNRPA1基因在胃癌中的表达情况。进一步构建SNRPA1慢病毒载体并转染到胃癌细胞系AGS中(shSNRPA1),采用CCK-8技术和克隆形成技术来分析SNRPA1对胃癌细胞生长增殖的影响。结果(1)在胃癌组织中,SNRPA1的表达水平明显高于正常组织,SNRPA1基因在胃癌等组织中呈现高表达(P<0.05);(2)通过细胞生物学实验发现,shSNRPA1组与阴性对照病毒组相比,shSNRPA1组的胃癌细胞AGS的增殖被抑制(P<0.05),由shSNRPA1组胃癌细胞AGS形成的可见集落数量明显少于阴性对照病毒组(P<0.05)。结论SNRPA1在胃癌中高表达;干扰SNRPA1可以抑制胃癌细胞AGS的生长增殖能力和集落形成能力。
Objective To observe the expression of SNRPA1 in gastric cancer and its effects on the growth of gastric cancer cells.Methods It was found through the analysis of High Content Screening(HCS)that interfering with SNRPA1 gene could inhibit the proliferation of AGS in gastric cancer cells.The expression level of SNRPA1 gene in gastric cancer was analyzed using TCGA database.SNRPA1 lentiviral vector was further constructed and transfected into gastric cancer cell line AGS(shSNRPA1).CCK-8 and clone formation were used to analyze the effects of SNRPA1 on the growth and proliferation of gastric cancer cells.Results(1)Bioinformatics analysis showed that the expression level of SNRPA1 in gastric cancer tissues was significantly higher than that in normal tissues.SNRPA1 gene was highly expressed in gastric cancer and other tissues(P<0.05);(2)It was found through the analysis of cell biology experiments that shSNRPA1 group could significantly inhibit the proliferation of gastric cancer cell line AGS(P<0.05)compared with the negative control virus group.The number of visible colonies formed by AGS in gastric cancer cells in shSNRPA1 group was significantly less than that in negative control virus group(P<0.05).Conclusion SNRPA1 is highly expressed in gastric cancer;interfering with SNRPA1 can inhibit the growth,proliferation and colony formation of AGS in gastric cancer cells.
