乙二胺二琥珀酸功能化硅胶材料的制备及特性研究

摘要 以乙二胺二琥珀酸(EDDS)为配体、3-缩水甘油基氧基丙基三甲氧基硅烷(γ-GLDP)为间隔臂、硅胶(Silica)为基质,制得一种新型多功能改性材料—EDDS-Silica。采用X射线光电子能谱(XPS)和酸碱电位滴定对EDDS-Silica分别进行了定性和定量表征,证明EDDS-Silica已成功制备,且当Silica与EDDS用量比为1.5∶3.0(m/m)时单位硅胶基质上EDDS的键合量最高,为39.82µmol/g。通过蛋白质色谱实验以及金属离子吸附实验分别对EDDS-Silica的阳离子交换特性和金属螯合特性进行了考察。结果表明,EDDS-Silica既可作为色谱固定相用于带电荷物质的分离与纯化,又可作为金属螯合吸附剂用于金属离子的吸附研究。EDDS-Silica优良的离子交换和金属螯合特性,使得该配体改性材料具有更为广阔的应用前景。 Ethylenediaminedisuccinic acid(EDDS)is a biodegradable amino polycarboxylic acid hexadentate compound containing six coordinating atoms.A multifunctional chromatographic material,EDDS-Silica,was developed by using EDDS as ligand,γ-glycidoxypropyltrimethoxysilane(γ-GLDP)as a spacer arm and silica as matrix.The prepared EDDS-Silica material was qualitatively characterized by X-ray photoelectron spectroscopy(XPS).Binding capacity of EDDS on the silica matrix was quantitatively characterized by acid-base potentiometric titration in order to optimize the best amount ratio of silica gel and EDDS.The results showed that the modified material was prepared successfully.When the amount ratio between silica and EDDS was 1.5∶3.0(m/m),the binding amount of EDDS on per unit silica matrix was the highest of 39.82µmol/g.Multifunction properties of the modified material including cation exchange behavior for proteins as well as metal chelating property for metal ions were investigated by protein chromatography and metalion adsorption experiments.The results showed that EDDS-Silica can be used not only as a chromatographic stationary phase for the separation and purification of charged substances,but also as a metal chelating adsorbent for the adsorption of metal ions.Multifunctional characteristics of EDDS-Silica involving excellent ion exchange and strong metal chelation make this ligand modified material have a wider application prospect.
