长期冻存对血小板聚集功能的影响

Effect of long-term cryopreservation on platelet aggregation

目的 探讨长期冻存对血小板聚集功能的影响。方法 使用冰冻血小板血液制剂和健康成人枸橼酸抗凝血样本建立冰冻血小板输注模型。瑞氏染色显微镜检血小板形态变化。血栓弹力图仪检测样本反应时间(R值)、凝固时间(K值)、Angle值、血栓最大幅度(MA值)。结果 与新鲜血小板相比,冰冻血小板形态不规则,边缘不整齐,直径明显增大且大小不均,聚集较多;血栓弹力图检测R值、K值、Ang值差异无统计学意义(P>0.05),MA值显著减小,差异有统计学意义(P<0.05)。结论 长期冻存使血小板活化和聚集功能降低。

Objective To investigate the effect of long-term cryopreservation on platelet aggregation.Methods Frozen platelet transfusion model was established by using frozen platelet blood preparation and citric acid anticoagulant samples of healthy adults.The morphological changes of platelets were examined by Wright staining microscope.The sample reaction time(R value),coagulation time(K value),Angle value and maximum thrombus amplitude(MA value)were detected by thromboelastograph.Results Compared with fresh platelets,frozen platelets had irregular shape,irregular edges,significantly larger diameter,uneven size and more aggregation;There was no significant difference in R value,K value and ang value by thromboelastography(P>0.05),but MA value decreased significantly(P<0.05).Conclusion Long term cryopreservation reduced platelet activation and aggregation.
