钙化性主动脉瓣膜疾病小鼠模型的构建

摘要 目的:本研究旨在探索有效快捷的钙化性主动脉瓣膜疾病小鼠模型的构建方式。方法:采用ApoE-/-背景小鼠、5/6肾切除以及高磷饲喂的方法建立小鼠钙化性主动脉瓣膜疾病的模型。分为假手术组(sham)9只,肾切除组(CKD)5只,造模后高磷饲喂4周。通过称量连续监测小鼠体质量,并在造模4周后,通过血尿素氮浓度评价小鼠肾功能减退情况,及检测血钙和血磷水平。对主动脉瓣以及主动脉弓部进行茜素红染色检测钙化程度。结果:与假手术组(sham)相比,肾切除组(CKD)小鼠体质量明显降低(P<0.05),血尿素氮浓度升高1.7倍(CKD vs.sham,P<0.05),血磷升高2.7倍(CKD vs.sham,P<0.05),提示小鼠肾功能减退。茜素红染色结果显示,造模4周后,肾切除组小鼠主动脉瓣根部和主动脉弓部即出现明显的钙沉积,提示小鼠钙化性主动脉瓣膜疾病模型造模成功。结论:ApoE-/-背景小鼠结合5/6肾切除以及高磷饲喂的方法可以快速有效建立小鼠钙化性主动脉瓣膜疾病的模型。 Objective:The purpose of this study was to explore the establishment of an effective and rapid mouse model of calcified aortic valve disease.ApoE-/-mice,5/6 nephrectomy and high phosphorus feeding were used to establish the model of calcified aortic valve disease in mice.They were divided into 10 mouse in sham operation group(sham)and 5 mouse in nephrectomy group(CKD).After 4 weeks of modeling,the weight of mice by weighing and the concentrations of blood calcium,blood phosphorus and blood urea nitrogen were measured to detect the renal function of mice and the calcium deposition was detected by alizarin red staining in aortic valve and aortic arch.The results showed that after 4 weeks of modeling,the weight of mice in CKD group was significantly lower than that in the sham group(P<0.05),and the concentrations of blood phosphorus were significantly 1.7 times higher than those in the sham group(CKD vs.sham,P<0.05)and the concentrations of blood urea nitrogen were significantly 2.7 times higher than those in the sham group(CKD vs.sham,P<0.05),suggesting renal failure.Alizarin red staining of aortic valve and aortic arch showed that there was obvious calcium deposition in aorta and aortic arch in the CKD group,suggesting that the model of calcified aortic valve disease in mice is successful.These results show that ApoE-/-mice combined with 5/6 nephrectomy and high phosphorus feeding is useful for effectively establishing the model of calcified aortic valve disease in mice.
