金线吊乌龟提取物的抗炎镇痛抗氧化和抑制亚硝化活性研究

摘要 目的:研究和比较金线吊乌龟Stephania cepharantha Hayata不同提取部位的抗炎、镇痛、抗氧化和抑制亚硝化活性。方法:采用二甲苯致小鼠耳廓肿胀法、醋酸扭体法和热板法考察金线吊乌龟不同提取部位的抗炎、镇痛活性;采用1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除法和2,2-联氮基-双-(3-乙基苯并噻唑啉-6-磺酸)二铵盐(ABTS)自由基清除法测定金线吊乌龟不同提取部位的抗氧化活性;采用盐酸萘乙二胺法和α-萘胺法测定金线吊乌龟不同提取部位的抑制亚硝化活性。结果:与空白组小鼠比较,水提取物对耳廓肿胀抑制率为36.57%,扭体抑制率为49.14%,且给药后1、2 h小鼠对热板的痛阈值分别提高47.40%和67.35%,其余各组差异均具有统计学意义(P<0.05);乙酸乙酯提取物清除DPPH自由基[半数抑制浓度(IC_(50))=(0.084±0.008)mg·mL^(–1)]和ABTS自由基[IC_(50)=(0.063±0.005)mg·mL^(–1)]的能力最强,并显著强于其他提取部位(P<0.05);水提取物清除亚硝酸盐[IC_(50)=(4.35±0.15)mg·mL^(–1)]和阻断亚硝胺[IC_(50)=(11.08±0.27)mg·mL^(–1)]的能力均著强于其他提取部位(P<0.05)。结论:金线吊乌龟不同提取部位均具有一定的抗炎、镇痛、抗氧化和抑制亚硝化活性,其中水提取物的抗炎、镇痛和抑制亚硝化活性最强,乙酸乙酯提取物的抗氧化活性最强。 Objective:To study and compare the activities of anti-inflammation,analgesia,antioxidation,and inhibiting nitrosation of different Stephania cephalantha Hayata extracts.Methods:The activities of anti-inflammation and analgesia of different S.cephalantha Hayata extracts were observed by the xylene-induced auricle swelling in mice,acetic acid writhing method,and hot plate method.The activity of antioxidation was evaluated by the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging and the 2,2’-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid,ABTS) radical scavenging methods.The activity of inhibiting nitrosation was determined by the methods of hydrochloric acid naphthalene ethyl acetateene diamine and α-naphthylamine.Results:Compared with the blank group,the inhibitory rates of water extract on xylene-induced auricle swelling and writhing response in mice were 36.57% and 49.14%.The pain threshold of mice on hot plates was increased by 47.40% and 67.35% at 1 hour and 2 hours after administration.The differences in other groups were all statistically significant(P<0.05).The ethyl acetate extract had the strongest DPPH [IC_(50)=(0.084±0.008) mg·mL^(–1)]and ABTS [IC_(50)=(0.063±0.005) mg·mL^(–1)] radical scavenging ability among other extracts(P<0.05).However,the ability of water extract to scavenge nitrite [IC(_(5)0)=(4.35±0.15) mg·mL^(–1)] and block nitrosamine [IC_(50)=(11.08±0.27) mg·mL^(–1)]was stronger than other extracts(P<0.05).Conclusion:Different S.cephalantha Hayata extracts had certain activities of antiinflammation,analgesia,antioxidation,and inhibiting nitrosation.Among the S.cephalantha Hayata extracts,the water extract had the strongest activities of anti-inflammation,analgesia,and inhibiting nitrosation,and the ethyl acetate extract had the strongest activity of antioxidation.
