摘要
为了确定PMEL基因在北极蓝狐和北极白狐不同组织中的表达特征,进而确定其与毛色形成的相关性,以北极蓝狐和北极白狐为试验对象,利用实时荧光定量PCR技术构建了前黑素小体蛋白基因(PMEL)在北极蓝狐和北极白狐的组织表达谱,并比较分析了该基因在以上2种不同毛色北极狐皮肤中mRNA的表达差异。结果表明,PMEL基因在北极蓝狐和北极白狐的不同组织(皮肤、心、肝、脾、肺、肾和肌肉)中均有表达,但在皮肤中的表达量均为最高,且北极蓝狐mRNA表达显著高于北极白狐(P<0.05),提示PMEL基因的表达与北极狐深色毛色的形成呈正相关。
Taking arctic blue fox and arctic white fox as the test subjects,the tissue expression profile of the premelanosome protein gene(PMEL)in arctic blue fox and arctic white fox was constructed using real-time fluorescent quantitative PCR technology,and the difference of mRNA expression of this gene in the skin of the these arctic foxes with different coat colors was analyzed to explore the expression characteristics of PMEL gene in different tissues of arctic blue fox and arctic white fox,and then to determine its correlation with the formation of coat color.The results showed that PMEL gene was expressed in different tissues(skin,heart,liver,spleen,lung,kidney and muscle)of arctic blue fox and arctic white fox,but the expression level in the skin was the highest,and arctic blue fox mRNA expression was significantly higher than that of arctic white fox(P<0.05),marking that PMEL gene expression was positively correlated with the formation of dark fur of arctic fox.
