金荞麦对急性溃疡性肠炎小鼠肠黏膜的保护作用及其机制的研究 被引量:3

摘要 目的:探金荞麦对溃疡性结肠炎(ulcerative colitis,UC)动物模型肠黏膜的保护作用及对IL-6/STAT3转导途径的调控作用。方法:将45只小鼠分为对照组、UC组、UC+金荞麦片组(每组15只)。利用3%的葡聚糖硫酸钠诱导UC模型。UC+金荞麦片组通过灌胃行金荞麦片干预。检测各组小鼠结肠长度、疾病活动指数、肠黏膜通透性、组织中炎症细胞因子水平、紧密连接蛋白表达水平以及IL-6、STAT3的转录和蛋白表达水平。结果:UC组疾病活动指数,IL-1β水平,TNF-α水平,肠黏膜通透性,IL-6、STAT3 mRNA和蛋白表达水平显著高于对照组(P<0.05),ZO-1、Occludin蛋白表达水平低于对照组(P<0.005),结肠长度显著短于对照组(P<0.05)。UC+金荞麦片组疾病活动指数,IL-1β水平,TNF-α水平、肠黏膜通透性,IL-6、STAT3 mRNA和蛋白表达水平显著低于UC组(P<0.05),ZO-1、Occludin蛋白表达水平高于UC组(P<0.05),结肠长度显著长于UC组(P<0.05)。结论:金荞麦可通过抑制IL-6/STAT3通路抑制炎症反应,保护肠黏膜屏障,缓解UC。 Objective:To explore the protective effect of Fagopyrum dibotrys on the animal model of ulcerative colitis(UC)and the regulation of IL-6/STAT3 transduction pathway.Methods:Forty-five mice were divided into control group,UC group,and UC+Fagopyrum dibotrys group(n=15).Use 3%dextransulfate sodium salt(DSS)was applied to induce UC model.Fagopyrum dibotrys was intervened by gavage.The colon length,disease activity index,intestinal mucosal permeability,inflammatory cytokine levels in tissues,tight junction protein expression levels,IL-6 and STAT3 transcription and protein expression levels were detected.Results:The disease activity index,IL-1β,TNF-α,intestinal mucosal permeability,IL-6 and STAT3 mRNA and protein levels in the UC group were significantly higher than those in the control group,ZO-1 and Occludin protein,and the colon length were significantly lower than those in the control Group(P<0.05).The disease activity index,IL-1β,TNF-α,intestinal mucosal permeability,IL-6 and STAT3 mRNA and protein levels in the UC+Fagopyrum dibotrys group were significantly lower than those in the UC group,ZO-1 and Occludin protein,and the colon length were significantly higher than those in the UC group(P<0.05).Conclusion:Fagopyrum dibotrys can inhibit IL-6/STAT3 pathway to inhibit inflammatory response,protect the intestinal mucosal barrier and relieve UC.
