摘要
碱性亮氨酸拉链核因子1(basic leucine zipper nuclear factor 1,BLZF1)属于b-ZIP核因子家族,编码一个3 kb的mRNA,可被翻译成45 kDa的蛋白质。BLZF1由1个亮氨酸重复序列和几个特定的磷酸化位点组成,其结构中包含亮氨酸拉链和核定位信号。BLZF1在人体正常细胞和肿瘤细胞中是普遍存在的;其在细胞核和细胞质中均有表达,但不在核仁中表达;但在类维生素A(retinoic acid,RA)治疗急性早幼粒细胞白血病(acute promyelocytic leukaemia,APL)引起细胞增殖时,其表达上调。BLZF1可与GRASP55结合,形成的结合体是一种新的高尔基体中池堆叠模型,在维持高尔基体正常结构和转运蛋白质中发挥重要作用。但其在多种疾病和肿瘤中的研究还未明确。本研究从BLZF1的结构、表达及功能方面,探讨其在一些疾病和肿瘤中的作用,为BLZF1在肿瘤中的研究提供理论基础。
BLZF1 (basic leucine zipper nuclear factor 1) belongs to the b-ZIP nuclear factor family and encodes a 3 kb mRNA translated into a 45 kDa protein.BLZF1 consists of a leucine repeat and several specific phosphorylation sites,and its structure contains a leucine zipper and a nuclear localization signal.BLZF1 is ubiquitous in human normal cells and tumor cells.It is expressed in the nucleus and cytoplasm,but not in the nucleolus.However,its expression is up-regulated when retinoic acid (RA) induces cell proliferation in APL (acute promyelocytic leukaemia).BLZF1 can bind to GRASP55,and the formed complex is a new model of pool stacking in the Golgi apparatus,which plays an important role in maintaining structure and transporting proteins in the Golgi apparatus.However,its research in various diseases and tumors has not been clear.In this study,we explored the role of BLZF1 in some diseases and tumors from the structure,expression and function of BLZF1,and provided a theoretical basis for the study of BLZF1 in tumors.
