抗乙肝病毒药物拉米夫定和恩替卡韦对OH2溶瘤效果的影响

Effects of Anti-hepatitis B Virus Drugs Lamivudine and Entecavir on the Oncolytic Effect of OH2

目的建立BALB/C小鼠CT26-iRFP结肠癌模型和评估药物拉米夫定(Lam)和恩替卡韦(Ente)对重组人GM-CSF溶瘤Ⅱ型单纯疱疹病毒(OH2)治疗肿瘤作用的影响。方法在18只4~6周龄雌性BALB/C小鼠右侧背部接种CT26-iRFP肿瘤细胞1×105个进行植瘤。随机分为4组,分别为OH2组、OH2+Lam组、OH2+Ente组、阴性对照组,每组4~5只小鼠。通过测量首次给药后的肿瘤体积来评估病毒溶瘤效果。结果成功在小鼠皮下种植CT26-iRFP模型,与阴性对照组相比所有注射OH2药物的动物肿瘤生长相对平缓。首次给药后21天,通过与阴性对照组比较,OH2组、OH2+Ente组对于CT26-iRFP肿瘤有明显效果(P<0.05),OH2+Lam组相比较阴性对照组无显著性差异(P=0.7536)。结论Ente对OH2治疗肿瘤效果无明显影响,腹腔注射OH2对小鼠皮下肿瘤有治疗效果,Lam对OH2溶瘤效果可能产生影响,但与阴性对照相比对肿瘤仍有抑制效果。

Objective To establish the CT26-iRFP colon cancer model in BALB/C mice,and to evaluate the effects of lamidine(Lam)and entecavir(Ente)on the treatment of tumor by recombinant human GM CSF oncolytic herpes simplex virus(OH2).Methods The CT26 iRFP model was successfully implanted subcutaneously in mice.Compared with the negative control group,the tumor growth of all animals injected with OH2 drugs was relatively gentle.21 days after the first administration,compared with the negative control group,the OH2 group and the OH2+Ente group had significant effects on CT26-iRFP tumor(P<0.05),while the OH2+Lam group had no significant difference compared with the negative control group(P=0.7536).Conclusion Ente has no obvious effect on tumor treatment with OH2.Intraperitoneal injection of OH2 has a therapeutic effect on subcutaneous tumor in mice.Lam may have an impact on the oncolytic effect of OH2,but it still has an inhibitory effect on tumor compared with the negative control.
