黄腐酚对人皮肤鳞癌A431细胞恶性生物学行为的抑制作用

摘要 目的探究黄腐酚对人皮肤鳞癌A431细胞增殖、迁移侵袭和凋亡的影响及作用机制。方法给予不同浓度梯度的黄腐酚(0、10、20、40、60、80、100μmol/L)处理A431细胞,应用四甲基偶氮唑盐(MTT)比色法检测细胞增殖活性;利用细胞划痕实验、Transwell小室迁移实验、Transwell小室侵袭实验检测黄腐酚处理后A431的迁移能力、侵袭能力;利用Western blot检测A431中STAT3、JAK2和Bcl-2蛋白的表达。结果黄腐酚可呈剂量依赖性抑制A431细胞的增殖、迁移和侵袭;A431细胞中STAT3、JAK2和Bcl-2有下调趋势,且均呈剂量依赖性。结论黄腐酚可呈剂量依赖性的抑制A431细胞的增殖和侵袭迁移,有望作为潜在的抗皮肤鳞癌药物应用到临床。 Objective To investigate the effect and mechanism of xanthohumol on the proliferation,migration,invasion,and apoptosis of human skin squamous cell carcinoma A431 cells.Methods A431 cells were treated with different concentration gradients of xanthohumol(0、10、20、40、60、80、100μmol/L),and the cell proliferation activity was detected by tetramethylazolium salt(MTT)colorimetric method.Cell scratch test,Transwell chamber migration test and Transwell chamber invasion test were used to detect the migration and invasion ability of A431 treated with xanthohumol.Western blot was used to detect the expression of STAT3,JAK2 and Bcl-2.Results Xanthohumol could inhibit the proliferation,migration and invasion of A431 cells in a dose-dependent manner.STAT3,JAK2 and Bcl-2 were down-regulated in a dose-dependent manner in A431 cells.Conclusion Xanthohumol inhibits the proliferation,invasion and migration of A431 cells,and promotes the apoptosis of A431 cells by regulating the expressions of STAT3,JAK2 and Bcl-2 in a dose-dependent manner.
