摘要
目的探讨锌指蛋白750(ZNF750)在结直肠癌细胞增殖中的作用和分子机制。方法用干扰siRNA转染结直肠癌细胞株SW620和LoVo,沉默ZNF750的表达;运用MTT实验和平板克隆实验检测ZNF750沉默成功的实验组细胞和对照组细胞增殖能力的差别;运用荧光定量PCR检测实验组细胞株中Kruppel样因子4(KLF4)的表达水平与对照组的差异。结果MTT实验和平板克隆实验显示ZNF750沉默后,结直肠癌细胞的增殖能力较对照组明显增强,差异具有统计学意义(P<0.05)。荧光定量PCR显示结直肠癌细胞株ZNF750沉默后,KLF4的表达水平下降,差异具有统计学意义(P<0.05)。结论ZNF750抑制结直肠癌细胞的增殖,可能通过调控KLF4的表达参与结直肠癌的发生发展。
Objective To investigate the role and of ZNF750 in colorectal cancer cell proliferation and its molecular mechanism.Methods siRNA was used to silence the expression of ZNF750 by transfecting into human CRC SW620 cells and LoVo cells.Then the success of transfection was detected by Real-time PCR.MTT assay and plate clone formation experiments were used to detect the proliferation activity of colorectal cancer cells between the ZNF750-silenced cells and Negative control cells.The mRNA expression levels of KLF4 in ZNF750-silenced cells and Negative control cells was detected by Real-time PCR.Results MTT results and plate clone formation experiments showed that ZNF750-silenced could enhance the proliferation activity of SW620 and LoVo cells(P<0.05).Real-time PCR showed that ZNF750-silenced could significantly decrease the mRNA expression levels of KLF4(P<0.05).Conclusion ZNF750 can inhibit the proliferation of colorectal cancer cells.ZNF750 may be involved by regulating the KLF4 to promote colorectal cancr cells.
