艾灸对慢性疲劳综合征大鼠肠黏膜屏障修复及疲劳改善作用的研究 被引量:6

摘要 目的:探究艾灸对慢性疲劳综合征(CFS)模型大鼠肠黏膜屏障结构及疲劳的改善作用。方法:将48只SD大鼠随机分成正常对照组、模型组、艾灸A组及艾灸B组,每组12只。正常组常规饲养,其余3组采用多因素诱导复合应激方法造模。模型组不加干预,艾灸A组给予温和灸神阙、关元穴,艾灸B组给予温和灸足三里穴(双侧),28 d后,评价大鼠一般情况,光镜下观察大鼠结肠组织形态学变化,采用Western blot和RT-PCR法检测结肠紧密连接ZO-1、Occludin蛋白和mRNA表达。结果:造模大鼠呈现反应迟钝、精神倦怠、活动减少、毛发枯槁、耳尾颜色苍白和便质稀软等表现。艾灸后好转,精神状态与排便情况改善较明显。光镜下可见模型组大鼠结肠黏膜充血水肿并有大量炎症细胞浸润,艾灸A、B组病理损伤明显减轻,杯状细胞数量增多。模型组ZO-1、Occludin蛋白与mRNA水平较正常组明显降低(P<0.01),艾灸组上述紧密连接蛋白相对表达均显著提升(P<0.01);且艾灸A组Occludin蛋白表达量高于艾灸B组(P<0.01)。结论:艾灸能减轻CFS模型大鼠结肠黏膜组织病变,上调紧密连接蛋白ZO-1、Occludin相对表达,保护肠黏膜屏障以抗疲劳;且艾灸神阙、关元穴稍优于艾灸足三里穴。 Objective:To explore the repair effect of Moxibustion on intestinal mucosal barrier in rats with chronic fatigue syndrome(CFS)and improvement on fatigue.Methods:SD rats were randomly divided into normal control group,model group,Moxibustion A and B groups,with 12 rats in each group.The normal group rats were fed routinely,and the other group rats were modeled with multi-factor-induced compound stress method for CFS model.The model group received no intervention,the Moxibustion A group was given mild moxibustion at Shenque and Guanyuan points,and the Moxibustion B group was given mild moxibustion at Zusanli(bilateral).After 28 days of treatment,the general conditions of rats were evaluated,the pathological changes of colon were observed under light microscope,and the expressions of ZO-1,occludin protein and mRNA in colonic tight junction were detected by Western blot and RT-PCR.Results:The CFS model rats showed signs of slow response,mental fatigue,reduced activity,hair withered,ear tail color pale,and soft skin,that recovered after moxibustion,and the mental state and defecation improved significantly.Under the light microscope,the colonic mucosa of rats in the model group was congested and edema with a large number of inflammatory cells infiltrated.The pathological changes in the Moxibustion A and B group were significantly alleviated,and the number of goblet cells was increased compared with the model group.The protein and mRNA levels of ZO-1 and Occludin in the model group were significantly lower than those in the normal group(P<0.01).After the intervention of moxibustion,the relative expressions of the above tight junction proteins were significantly increased(P<0.01).The expression of Occludin protein in Moxibustion A group was higher than that in Moxibustion B group(P<0.01).Conclusion:Moxibustion can repair the colonic mucosa pathological changes in CFS model rats,up-regulate the relative expressions of tight junction protein ZO-1,occludin,and protect intestinal mucosal barrier against fatigue.Effect of moxibustion at Shenque and Guanyuan is slightly better than that of moxibustion at Zusanli.
