响应面法优化黄花菜黄酮提取工艺及抗氧化活性的研究 被引量:11

Optimization of Extraction Process of Flavonoids from Hemerocallis citrina Baroni by Response Surface Methodology and Study on Antioxidant Activity

摘要 以黄花菜黄酮提取量为考察指标,通过单因素实验结合响应面分析的方法,优化黄花菜黄酮索氏提取法提取工艺,并采用清除DPPH和ABTS自由基法测定其抗氧化活性。结果表明,黄花菜黄酮索氏提取最佳工艺条件为:提取时间2.5 h,乙醇浓度90%,料液比1∶20(g/mL),该工艺条件下,黄花菜黄酮提取量约为0.55%,相对标准偏差为2.11%。黄花菜黄酮与V_(C)清除DPPH自由基的IC_(50)分别为0.056 mg/mL和0.021 mg/mL,清除ABTS自由基的IC_(50)分别为0.056 mg/mL和0.031 mg/mL。表明响应面优化黄花菜黄酮索氏提取工艺准确、可靠、稳定,且黄花菜黄酮具有较强的抗氧化活性,为黄花菜黄酮等成分的提取及综合利用提供了参考依据。 The Soxhlet extraction process of flavonoids from Hemerocallis citrina Baroni is optimized with flavonoid yield as the index by single-factor experiment combined with response surface analysis.The antioxidant activity of flavonoids is determined by DPPH and ABTS free radicals scavenging method.The results show that the optimal extraction conditions for Soxhlet extraction of flavonoids from Hemerocallis citrina Baroni are as follows:extraction time is 2.5 hours,ethanol concentration is 90%,and the ratio of solid to liquid is 1∶20(g/mL).Under such conditions,the yield of flavonoids from Hemerocallis citrina Baroni is about 0.55%,and the relative standard deviation is 2.11%.The IC_(50) values of flavonoids from Hemerocallis citrina Baroni and Vc scavenging DPPH free radicals are 0.056 mg/mL and 0.021 mg/mL respectively;and the IC_(50) values of flavonoids from Hemerocallis citrina Baroni and Vc scavenging ABTS free radicals are 0.056 mg/mL and 0.031 mg/mL respectively,which indicates that the Soxhlet extraction process of flavonoids from Hemerocallis citrina Baroni optimized by RSM is accurate,reliable and stable,and the flavonoids from Hemerocallis citrina Baroni have strong antioxidant activity,providing a reference basis for the extraction and comprehensive utilization of flavonoids from Hemerocallis citrina Baroni.
