基于网络药理学探讨茵陈四逆汤治疗慢加急性肝衰竭的分子机制

Molecular mechanism research of Yinchen Sini decoction in treating acute-on-chronic liver failure based on network pharmacology

目的:通过网络药理学和分子对接的方法,初步探讨茵陈四逆汤治疗慢加急性肝衰竭的潜在机制。方法:从TCMSP数据库收集茵陈四逆汤中各药物的成分和作用靶点;Genecards、OMIM和TTD数据库收集慢加急性肝衰竭相关靶点;STRING数据库构建蛋白相互作用网络,Metascape数据库进行基因本体(GO)分析和基因组百科全书(KEGG)分析,以明确其参与重要生物过程及通路。结果:共筛选出131个茵陈四逆汤中的药物活性成分与236个治疗慢加急性肝衰竭的治疗靶点。PPI网络分析茵陈四逆汤治疗慢加急性肝衰竭可能与JUN、TNF、IL1B、IL6、STAT3、IL4、CXCL8、CCL2、MAPK3、RELA等蛋白有关,GO和KEGG结果显示茵陈四逆汤治疗慢加急性肝衰竭的靶点显著富集于AGEs-RAGE通路、IL-17信号通路、TNF信号通路、PI3K-Akt信号通路、HIF-1信号等通路。结论:网络药理学初步探讨了茵陈四逆汤治疗慢加急性肝衰竭的作用机制,为慢加急性肝衰竭治疗提供新思路。

Objective:To predict the potential targets and mechanisms of Yinchen Sini decoction in the treatment of acute-on-chronic liver failure based on network pharmacology.Methods:TCMSP database, GeneCards database, OMIM database and TTD database were used to predict and screen the active constituents and therapeutic targets of Yinchen Sini decoction.The protein-protein interaction(PPI) was analyzed by String database.To identify the important biological processes and pathways involved, the Metascape database was used for GO enrichment analysis and KEGG pathway enrichment. Results:In this study, 131 active components and 236 therapeutic targets were screened. PPI network analysis showed that Yinchen Sini Decoction might be related to Jun, TNF, IL1 B, IL6, STAT3, IL4, CXCL8, CCL2,MAPK3,RELA and other proteins. Go and KEGG results showed that the targets of Yinchen Sini Decoction in the treatment of acute-on-chronic liver failure were significantly enriched in AGEs-RAGE pathway, IL-17 signaling pathway, TNF signaling pathway, PI3 K-Akt signaling pathway, HIF-1 signaling pathway. Conclusion:The mechanism of Yinchen Sini Decoction in the treatment of acute-on-chronic liver failure was preliminarily discussed in network pharmacology, which provided new ideas for the treatment of chronic and acute liver failure.
