肝癌组织TPX2、TUFT1、Vimentin表达与癌细胞恶性生物学行为的关系及预测远处转移的价值研究

The relationship between the expression of TPX2 protein, TUFT1 and vimentin in liver cancer tissues and the malignant biological behavior of cancer cells and the value of predicting distant metastasis

目的:探讨肝癌组织爪蟾驱动蛋白样蛋白2靶蛋白(TPX2)、釉丛蛋白1(TUFT1)、波形蛋白(Vimentin)表达与癌细胞恶性生物学行为的关系及预测远处转移的价值。方法:选取2018年1月~2020年4月收治的51例肝癌远处转移患者为观察组,51例肝癌无远处转移患者为对照组,比较两组患者一般资料、TPX2 mRNA、TUFT1和Vimentin蛋白、癌细胞恶性生物学行为指标[B淋巴细胞瘤-2(Bcl-2)mRNA、survivin mRNA、Bax mRNA、半胱氨酸天冬氨酸蛋白酶-3(caspase-3)mRNA],分析TPX2 mRNA、TUFT1和Vimentin蛋白之间及与癌细胞恶性生物学行为指标、病理特征的相关性,并分析TPX2 mRNA、TUFT1和Vimentin蛋白预测远处转移的价值。结果:观察组患者TPX2 mRNA、TUFT1和Vimentin蛋白表达高于对照组患者(P<0.05)。TPX2 mRNA与TUFT1、Vimentin蛋白呈正相关,TUFT1蛋白与Vimentin蛋白呈正相关(P<0.05)。TPX2 mRNA、TUFT1蛋白、Vimentin蛋白与Bcl-2 mRNA、survivin mRNA均呈正相关,与Bax mRNA、caspase-3 mRNA均呈负相关(P<0.05)。TPX2 mRNA、TUFT1蛋白、Vimentin蛋白与肿瘤T、N分期呈正相关,与分化程度呈负相关(P<0.05)。TPX2 mRNA、TUFT1、Vimentin联合预测肝癌远处转移的AUC最大,为0.932。结论:肝癌组织TPX2、TUFT1、Vimentin表达与癌细胞恶性生物学行为存在一定相关性,早期采用三者联合检测方式,有望成为临床预测肝癌远处转移、制定恰当治疗方案的新途径。

Objective: To investigate the relationship between the expression of targeting protein for Xenopus kinesin-like protein 2( TPX2),Tuftelin 1( TUFT1),Vimentin in liver cancer tissues and the malignant biological behavior of cancer cells and the value of predicting distant metastasis. Methods: From January 2018 to April 2020,51 patients with distant metastasis of liver cancer( observation group,Tx Nx M1) and 51 patients with liver cancer without distant metastasis( control group,Tx Nx M0) admitted to our hospital were selected. The general data,TPX2 m RNA,TUFT1,Vimentin,and malignant biological behavior indicators of cancer cells [B lymphocyte tumor-2( Bcl-2)m RNA,survivin m RNA,Bax m RNA,caspase-3 m RNA] were compared between the two groups. The correlation between TPX2 m RNA,TUFT1,Vimentin and the malignant biological behavior indicators and pathological characteristics of cancer cells were analyzed,and the value of TPX2 m RNA,TUFT1,Vimentin in predicting distant metastasis was analyzed. Results: The expressions of TPX2 m RNA,TUFT1 and Vimentin in the observation group were higher than those in the control group( P < 0. 05). TPX2 m RNA was positively correlated with TUFT1and Vimentin,and TUFT1 was positively correlated with Vimentin( P < 0. 05). TPX2 m RNA,TUFT1 and Vimentin were positively correlated with Bcl-2 m RNA and survivin m RNA,and negatively correlated with Bax m RNA and caspase-3 m RNA( P < 0. 05). TPX2 m RNA,TUFT1 and Vimentin were positively correlated with tumor T and N stages,and negatively correlated with the degree of differentiation( P <0. 05). The AUC of TPX2 m RNA,TUFT1 and Vimentin in the joint prediction of distant metastasis of liver cancer was the largest,which was 0. 932. Conclusion: The expressions of TPX2 protein,TUFT1 and Vimentin in liver cancer tissues are related to the malignant biological behavior of cancer cells. The early use of the combined detection method of the three is expected to become a new way to predict the distant metastasis of liver cancer in the clinic and formulate an appropriate treatment plan.
