摘要
目的:探究不同菌种(G^(+)与G^(-))所致脓毒症诱导大鼠心肌组织匀浆一氧化氮(NO)水平与时间的关系。方法:选取105只健康SD雄鼠,随机分为革兰阳性细菌脓毒症组(G^(+)组,n=45)、革兰阴性细菌脓毒症组(G^(-)组,n=45)、生理盐水对照组(NS组,n=45),分别予以腹腔注射热灭活金黄色葡萄球菌(7.35×10^(12) CFU/kg)、热灭活大肠埃希菌(1.26×10^(12) CFU/kg)、生理盐水,注射体积均为3 ml。造模成功后,于第7天、第14天、第28天分别留取大鼠心肌组织,采用Elisa试剂盒检测组织匀浆中一氧化氮(NO)的浓度。结果:与NS组相比,G^(+)组及G^(-)组大鼠心肌组织匀浆NO浓度于7 d开始升高,14 d左右达到峰值,此后NO浓度开始下降,差异有统计学意义(P<0.05)。其中G^(+)组及G^(-)组之间各检测时间点差异无统计学意义(P>0.05)。结论:NO可能参与脓毒症心肌病的发生发展过程,其在革兰阳性菌与革兰阴性菌所致脓毒症大鼠心肌损伤之间表达差异不大。
Objective To explore the relationship between nitric oxide(NO)level and time in myocardial tissue of rats with sepsis induced by different strains(G^(+)and G^(-)).Method A total of 105 healthy SD male rats were randomly divided into Gram-positive bacterial sepsis group(G^(+)group),Gram-negative bacterial sepsis group(G^(-)group),and normal saline control group(NS group).The rats were injected with intraperitoneal injection of heat-inactivated Staphylococcus aureus(7.35×10^(12) CFU/kg),heat-inactivated Escherichia coli(1.26×10^(12) CFU/kg),and normal saline,with the injection volume of 3 ml.After successful modeling,the myocardial tissues of rats were collected on the 7 th,14 th and 28 th day,and the concentration of nitric oxide in tissue homogenate was detected by Elisa kit.Results Compared with the NS group,the serum NO concentration in the G^(+)and G^(-)groups began to increase at 7 d,and reached the peak at about 14 d,then the NO concentration began to decrease,the difference was statistically significant(P<0.05).There was no significant difference in the detection time points between the G^(+)and G^(-)groups(P>0.05).Conclusion NO may be involved in the occurrence and development of septic cardiomyopathy.There is little difference in the expression of NO between Gram-positive and Gram-negative bacteria-induced myocardial injury in septic rats.
