摘要
动脉粥样硬化(atherosclerosis,AS)是脂质在动脉内膜异常沉积所致的慢性炎症性疾病。NLRP3炎症小体在脂质代谢紊乱等AS常规危险因素和体细胞突变等AS新危险因素的形成机制中发挥极其重要的作用,并成为AS的生物学标志。该文主要概括了NLRP3炎症小体在胆固醇及老化等因素介导的动脉粥样硬化中的作用及机制,以及以NLRP3炎症小体作为靶点治疗AS的现状,为治疗冠心病及中风等疾病提供新策略。
Atherosclerosis(AS)is a chronic inflammatory disease caused by abnormal deposition of lipid in the intima of the artery.The NLRP3 inflammasome plays a significant role in the pathogenesis of conventional risk factors such as lipid metabolism disorder and newly-found risk factors such as somatic mutation,which has become a biomarker of AS.This review summarized the role and mechanisms of action of NLRP3 inflammasome on atherosclerosis mediated by cholesterol and aging.The potential value of NLRP3 inflammasome as a therapeutic target is also discussed.Exploring the mechanism of NLRP3 inflammasome in regulating AS will help to develop new therapeutic strategies for the treatment of coronary heart disease and stroke.
