姜黄素磷脂固体分散体对高脂饮食诱导的动脉粥样硬化的保护作用研究 被引量:1

Study on the Protective Effect of Curcumin Phospholipid Solid Dispersion on High Fat Diet Induced Arteriosclerosis

摘要 探讨姜黄素磷脂固体分散体对高脂饮食诱导兔动脉粥样硬化的影响。采用单纯高脂饲料喂养法制备动脉粥样硬化(Atherosclerosis,AS)模型;随机分成正常组、模型组、阳性组、姜黄素组、姜黄素磷脂固体分散体组,每组6只。药物干预组单剂量给药,灌胃口服100 mg/(kg·d)(以纯姜黄素计)。阳性组给予阿托伐他汀钙1.5 mg/(kg·d)(以纯阿托伐他汀钙计)。正常组、模型组给予等计量的生理盐水。8 w后检测各组血脂水平:HDL-C、LDL-C、TG和TC;炎性因子及抗氧化能力:IL-6、hs-CRP、MCP-1、TNF-α、T-AOC、SOD、CAT、MDA、ET等指标;并作主动脉粥样硬化病理检测。与阳性组和姜黄素组相比,姜黄素磷脂固体分散体组血脂检测LDL-C、TG及TC含量均显著降低,HDL-C含量显著升高,差异具有统计学意义(P<0.05);抗氧化检测T-AOC、SOD、CAT水平均显著升高,MDA水平显著降低,差异具有统计学意义(P<0.05);抗炎因子检测TNF-α、IL-6、hs-CRP、MCP-1、ET因子水平降低,差异具有统计学意义(P<0.05);血管组织病理学评价内膜与中膜厚度比值明显减小,斑块缩小明显,泡沫细胞和巨噬细胞少而散在。姜黄素磷脂固体分散体在胆固醇调节、抗氧应激反应、抑制炎症因子表达、维持细胞功能等AS治疗靶点的调节方面都有显著的效果,明显高于阳性对照组。 To study the effect of curcumin phospholipid solid dispersion on arteriosclerosis in rabbits induced by high fat diet,the AS models were prepared by high fat diet,and randomly divided into normal group,model group,positive group,curcumin group,curcumin phospholipid solid dispersion group,6 mice in each group.The drug intervention group was orally given a single dose of 100 mg/(kg·d)(calculated as pure curcumin).The positive group was given atorvastatin calcium 1.5 mg/(kg·d)(as pure atorvastatin calcium).Normal group and model group were given equal amount of normal saline.After 8 weeks,the blood lipid levels of each group were measured:HDL-C,LDL-C,TG and TC;Inflammatory factors and antioxidant capacity:IL-6,hs-CRP,MCP-1,TNF-α,T-AOC,SOD,CAT,MDA,ET indexes.Aortic atherosclerosis was also detected by pathology.Compared with positive group and curcumin group,the levels of LDL-C,TG and TC in curcumin phospholipid solid dispersion group were significantly lower,and the levels of HDL-C were significantly higher(P<0.05).In CPG group,the levels of T-AOC,SOD and CAT were significantly increased(P<0.05),while the levels of MDA,TNF-α,IL-6,hs-CRP,MCP-1 and ET were significantly decreased,the difference was statistically significant(P<0.05),the ratio of intima-to-media thickness(IMT)of vascular histopathology was significantly decreased,the plaque was small,the foam cells and macrophages were few and scattered.Curcumin phospholipid solid dispersion has a significant effect on cholesterol regulation,anti-oxidative stress response,inhibition of inflammatory factors expression,maintenance of cell function and other targets of AS treatment,which has significantly higher effect than that of positive control.
