摘要
目的探讨高同型半胱氨酸血症与大鼠氧化应激及海马形态学改变的相关性方法应用L-半胱氨酸制备大鼠高同型半胱氨酸血症模型,8周后,取血、脑、分离大鼠海马。检测血清及脑组织中Hcy、MDA、SOD的含量,脑组织Caspase-3水平及光镜下海马形态学的改变。结果大鼠模型组血清和脑组织中的Hcy、MDA含量明显高于空白对照组组(P<0.01),SOD含量明显低于空白对照组组(P<0.01);Caspase-3水平明显高于空白对照组(P<0.01);模型组大鼠海马光镜下神经细胞稀疏,密度下降,排列不规则,微血管周围间隙增宽,神经纤维网疏松,神经元胞体肿胀、水肿,呈海绵状变性改变。结论高同型半胱氨酸血症大鼠通过氧化应激及凋亡损伤海马。
Objective To investigate the correlation between hyperhomocysteinemia and oxidative stress and hippocampal morphological changes in rats.Methods L-cysteine was used to establish a rat model of hyperhomocysteinemia.After 8 weeks,the blood,brain and hippocampus were isolated.The contents of Hcy,MDA and SOD in serum and brain tissue,the level of Caspase-3 in brain tissue and the changes of hippocampus morphology under light microscope were detected.Results The contents of Hcy and MDA in the serum and brain tissue of the model group were significantly higher than those of the blank control group(P<0.01),and the content of SOD was lower than that of the blank control group(P<0.01).The level of Caspase-3 was significantly higher than that of the blank control group(P<0.01).The hippocampus of rats in the model group had sparse nerve cells,decreased density,irregular arrangement,widened space around capillaries,loose nerve fiber network,swelling and edema of neuron cell bodies,showing spongiform degeneration under the optical microscope.Conclusion The hippocampus of hyperhomocysteinemia rats was damaged by oxidative stress and apoptosis.
