SREBP1、ABCA1在颈动脉狭窄患者中的表达及意义

Expressions of SREBP1 and ABCA1 in patients with carotid artery stenosis and their significance

目的观察颈动脉狭窄患者的固醇调节元件结合蛋白(SREBP1)、腺苷三磷酸结合盒转运体A1(ABCA1)表达水平,分析SREBP1、ABCA1与颈动脉狭窄严重程度的关系。方法62例颈动脉狭窄患者中狭窄程度<70%(A组)37例和≥70%(B组)25例,检测患者的血脂、炎症因子、SREBP1、ABCA1水平,并与健康体检者(C组)对比,分析SREBP1、ABCA1与血脂、炎症因子之间的相关性。结果3组的总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、SREBP1比较,均为B组>A组>C组,3组高密度脂蛋白(HDL-C)、ABCA1水平比较,均为B组<A组<C组,差异均有统计学意义(P<0.05)。颈动脉狭窄患者的SREBP1水平与TC、TG、LDL-C、TNF-α、IL-6、IL-1β呈正相关,与HDL-C呈负相关;ABCA1水平与TC、TG、LDL-C、TNF-α、IL-6、IL-1β呈负相关,与HDL-C呈正相关,均P<0.05。结论脂质代谢紊乱、炎症反应与颈动脉狭窄严重程度密切相关,其原因可能与患者血清中SREBP1上调及ABCA1下调有关。

Objective To observe the expression levels of sterol regulatory element binding protein-1(SREBP1)and adenosine triphosphate binding cassette transporter A1(ABCA1)in patients with carotid stenosis,and to analyze the relationship between SREBP1 and ABCA1 and the severity of carotid stenosis.Methods Among 62 patients with carotid artery stenosis,37 had stenosis degree<70%(group A),and 25 had stenosis degree≥70%(group B).The levels of blood lipids,inflammatory factors,SREBP1 and ABCA1 were measured,and were compared with healthy controls(group C).The correlations between SREBP1,ABCA1 and blood lipids and inflammatory factors were analyzed.Results The levels of total cholesterol(TC),triglyceride(TG),low-density lipoprotein(LDL-C),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β)and SREBP1 in the three groups were all in the order of group B>group A>group C.The levels of high-density lipoprotein(HDL-C)and ABCA1 in the three groups were all in the order of group B<group A<group C.The differences were all statistically significant(P<0.05).The SREBP1 level of patients with carotid artery stenosis was positively correlated with TC,TG,LDL-C,TNF-α,IL-6 and IL-1β,and was negatively correlated with HDL-C.The ABCA1 level was negatively correlated with TC,TG,LDL-C,TNF-α,IL-6 and IL-1β,and was positively correlated with HDL-C.All P<0.05.Conclusions Lipid metabolism disorder and inflammatory reaction are closely related to the severity of carotid artery stenosis,which may be related to up-regulation of SREBP1 and down-regulation of ABCA1 in patients’serum.
