摘要
目的探讨p53基因突变和p16基因缺失与肝癌的相关性。方法采用黄冈市中心医院肿瘤科肝癌组织标本30例。运用饱和酚-氯仿法,提取DNA。根据Gen Bank数据或其他参考文献,设计出1对p53基因外显子8的PCR引物。进行PCR扩增,观察目标条带,将PCR产物进行纯化、测序、分析。免疫组织化学方法SP-p16,对其突变和缺失进行分析。结果在近30名肝癌患者体检中,有15名检测到血浆循环核酸p53突变。有癌转移史的病人17例突变率大大高于癌未发转移前13例。结论p53基因突变和血浆p16基因编码缺陷都与肝癌的发生、严重化程度加重和癌转移发生有关。
Purpose To investigate the correlation between p53 gene mutation and p16 gene deletion and hepatocellular carcinoma.Methods Thirty cases of liver cancer tissue samples were collected from Huanggang Central Hospital.DNA was extracted using the method of saturated phenol-chloroform.According to GenBank data or other references,a PCR primer for exon 8 of p53 gene was designed.PCR amplification was performed,the target bands were observed,and the PCR products were purified,sequenced and analyzed.Consequence In nearly 30patients with liver cancer check-up,plasma circulating nucleic acid was detected in 15 of p53 mutations.17 patients with a history of cancer metastasis mutation rate much higher than 13 cases before carcinoma without metastasis.Conclusion Both p53 gene mutation and plasma p16 gene coding defect are associated with the occurrence,severity and metastasis of HCC.
