摘要
为探究灭活的羊口疮病毒(ORFV)调节宿主免疫应答后对口蹄疫病毒(FMDV)复制的调控作用,本研究通过制备、灭活、纯化ORFV,采用RT-qPCR方法评估并分析在预先使用灭活的ORFV处理的BHK-21细胞中FMDV的复制情况以及细胞因子的变化情况。研究结果显示,灭活的ORFV上调细胞因子GM-CSF、IFN-γ、IL-1α、IL-2、IL-6和IL-10的转录水平,显著抑制FMDV在BHK-21细胞中的复制。也就是说,灭活的ORFV通过上调宿主细胞因子的转录进而抑制FMDV复制。这一研究结果为灭活的ORFV作为免疫调节剂提供了依据并为其应用提供了可能性。
The study aims to explore the regulatory effect of inactivated orf virus(ORFV)on the replication of foot-and-mouth disease virus(FMDV)after the host immune response regulated by ORFV.In this study,replication of FMDV and the regulation of cytokines in BHK-21 cells treated with inactivated ORFV were evaluated and analyzed by RT-q PCR.In result,FMDV was inhibited in BHK-21 cells and cytokines GM-CSF,IFN-γ,IL-1α,IL-2,IL-6 and IL-10 were up-regulated by the inactivated ORFV.In a word,the inactivated ORFV inhibits FMDV replication by up regulating the transcription of host cytokines.This study result provided the theoretical base that the inactivated ORFV may play the role of immune regulator and supplied the possibility for its application.
