RASSF1基因表达水平与肝癌的相关性及对细胞生理活动的作用

摘要 目的探究RASSF1基因表达水平与肝癌的相关性及对细胞生理活动的作用。方法选取154份肝癌病理组织样本,通过逆转录-聚合酶链式反应(RT-PCR)技术、CCK-8实验、transwell迁移、侵袭实验以及记录患者预后状况等方法明确RASSF1对于肝癌细胞生理活动的作用。结果肝癌组织中RASSF1表达水平低于癌旁组织。RASSF1基因表达水平与肝癌肿瘤大小、病理分级、AJCC临床分期相关(P<0.05),与患者的年龄、性别无明显相关性(P>0.05)。低表达RASSF1的肝癌患者预后较差,而高表达RASSF1的肝癌患者普遍预后较好。表明RASSF1表达水平与患者的预后存在明显相关。低表达细胞增殖OD值为(1.45±0.19),高于对照细胞的(0.97±0.09),差异有统计学意义(P<0.05)。迁移实验中,低表达细胞穿膜细胞数为(683±45),高于对照细胞的(384±29),差异有统计学意义(P<0.05)。侵袭实验中,低表达细胞穿膜细胞数为(512±33),高于对照细胞的(291±12),差异有统计学意义(P<0.05)。证明低表达RASSF1可以促使细胞增殖、迁移、侵袭能力增强。结论RASSF1具有成为肝癌早期诊断治疗的分子生物学标志的潜在价值。RASSF1基因或将有可能成为临床诊治肝癌的潜在分子作用靶点。 Objective To investigate the correlation between RASSF1 gene expression and hepatocellular carcinoma and its effect on cellular physiological activities.Methods 154 hepatocellular carcinoma pathological tissue samples were selected,and the role of RASSF1 on the physiological activity of hepatocellular carcinoma cells was clarified by reverse transcriptase-polymerase chain reaction(RT-PCR)technique,CCK-8 assay,transwell migration,invasion assay and recording the prognostic status of patients.Results The expression level of RASSF1 in hepatocellular carcinoma tissues was lower than that in paraneoplastic tissues.RASSF1 gene expression level correlated with tumor size,pathological grade,and clinical stage of AJCC in hepatocellular carcinoma(P<0.05),but had no significant correlation with the age and gender of the patients(P>0.05).Patients with low expression of RASSF1 had a poor prognosis,while those with high expression of RASSF1 generally had a better prognosis.It showed that there was a significant correlation between the expression level of RASSF1 and the prognosis of patients.The OD value of cell proliferation in the low expression cells was(1.45±0.19),which was higher than(0.97±0.09)in the control cells,and the difference was statistically significant(P<0.05).In the migration assay,the invasive cell number in the low expression cells was(683±45),which was higher than(384±29)in the control cells,and the difference was statistically significant(P<0.05).In the invasion assay,the invasive cell number in the low expression cells was(512±33),which was higher than(291±12)in the control cells,and the difference was statistically significant(P<0.05).It was proved that low expression of RASSF1 can promote cell proliferation,migration and invasion.Conclusion RASSF1 has potential value as a molecular biological marker for early diagnosis and treatment of hepatocellular carcinoma,and may become a potential molecular target for clinical diagnosis and treatment of hepatocellular carcinoma.
