角叉菜粗多酚的提取及抑癌活性研究

Extraction of Crude Polyphenols from Chondrus Ocellatus and their Antitumor Activity

目的:研究角叉菜中粗多酚提取工艺及其抗癌活性。方法:采用超声辅助乙醇提取,福林酚法测定含量,并分别采用苯酚硫酸法、考马斯亮蓝G250法、灼烧法和酸碱处理法测定粗多酚中的多糖、蛋白质、总灰分和粗纤维成分。用CCK-8细胞活力测定法测定角叉菜粗多酚对人肺癌A549和H1975细胞活力的影响。结果:乙醇体积分数为61%、超声时间为41 min,超声温度为60℃时,多酚得率为24.8%,多糖6.94%,蛋白质17.1%,总灰分33.4%,粗纤维1.83%。结论:角叉菜粗多酚主要组分对人肺癌细胞活力具有一定抑制作用。

Objective:To study the extraction process of crude polyphenols from Chondrus ocellatus and their anticancer activity.Methods:Polyphenols were extracted from Chondrus ocellatus using ultrasonic-assisted ethanol method,and their contents were determined by Folin-Ciocalteu method.Respectively,the polysaccharides,protein,total ash and crude fiber components of crude polyphenols were detected by phenol sulfuric acid method,Coomassie brilliant blue G250 method,burning and acid-base treatment.The CCK-8 cell viability assay detected the effects of crude polyphenols on the human lung cancer A549 and H1975.Results:Under the optimized conditions(ethanol concentration,61%;ultrasound time,41min;ultrasound temperature,60℃),the yield of polyphenols,polysaccharides,protein,total ash and crude fiber were 24.8%,6.94%,17.1%,33.4 %and 1.83%,respectively.Conclusion:The main components of crude polyphenols from Chondrus ocellatus can inhibit the viability of human lung cancer cells to a certain extent.
