摘要
采用在线高效液相色谱-1,1-二苯基-2-三硝基苯肼(在线HPLC-DPPH)活性筛选系统,结合中、高压反相液相色谱,定向分离沙棘中自由基抑制剂.该方法可以实现从沙棘复杂成分中对目标成分进行富集并快速识别活性成分.通过高压反相液相色谱对目标峰进行分离纯化,并利用ESI-MS和NMR数据对化合物进行结构鉴定.结果表明,该方法能够快速、高效地从沙棘中识别分离纯化自由基抑制剂,为进一步进行药理活性研究和从天然产物中快速制备具有抗氧化活性黄酮类化合物提供参考.
By using the online HPLC-DPPH activity screening system which was combined with medium and high pressure reversed-phase liquid chromatography,free radical inhibitors was directly isolated from Sea buckthorn.The method ccould enrich the target components from the complex components of sea buckthorn and quickly identify the active components.The target peaks can be separated and purified by high-pressure reversed-phase liquid chromatography.ESI-MS and NMR data was applied to identify the structures of compounds.The results showed that this method can quickly identify and purify free radical inhibitors from sea buckthorn,which provided a reference for further study of pharmacological activity and rapid preparation of flavonoids with antioxidant activity from natural products.
