摘要
兔出血症病毒(RHDV)是兔出血症的病原体,可导致成年兔大量死亡.研究表明,细胞因子在RHD致病中发挥了重要作用.为探索细胞因子在RHDV对幼兔和成年兔致病差异性中的作用机制,通过对幼兔(2周龄)和成年兔(3月龄)肌肉注射检测为RHDV核酸阳性的病兔肝脏悬液,酶联免疫吸附试验(ELISA)检测感染RHDV后不同时间(0、12、24、36、48、60 h)血清中IL-8、IL-10、TNF-α、TGF-β和MCP-1的水平.ELISA结果显示,成年兔感染RHDV后血清中IL-8、IL-10、TNF-α、TGF-β和MCP-1水平在0~36 h显著升高(P<0.05),并在感染后36~48 h死亡.幼兔感染后血清中IL-8水平在24~60 h明显上升(P<0.05),TNF-α和TGF-β水平变化无明显规律,IL-10和MCP-1水平无明显变化,且未出现死亡现象.比较幼兔和成年兔上述细胞因子水平发现,成年兔感染后血清中IL-8、IL-10、TGF-β和MCP-1水平显著高于幼兔(P<0.05),TNF-α水平在感染后24 h左右高于幼兔(P<0.001),其余时间均低于幼兔(P<0.01).研究结果提示,IL-8、IL-10、TNF-α、TGF-β和MCP-1在血清中的水平差异可能是影响RHDV对幼兔与成年兔致病差异的关键因素.
Rabbit hemorrhagic disease virus(RHDV) is the pathogen of rabbit hemorrhagic disease,which causes massive deaths in adult rabbits.Studies have shown that the cytokines played an important role in the pathogenesis of RHD.To explore pathogenic mechanism and the difference of cytokines in RHDV of immature rabbit in the adult rabbit,the liver suspensions of young rabbits(2 weeks old) and adult rabbits(3 months old) were injected intramuscularily with the positive for RHDV nucleic acid.Enzyme-linked immunosorbent assay(ELISA) was utilized to determine serum levels of IL-8,IL-10,TNF-α,TGF-β and MCP-1 at the different times(0,12,24,36,48,60 h) after infection of RHDV.ELISA results showed that the serum levels of IL-8,IL-10,TNF-α,TGF-β and McP-1 in adult rabbits infected with RHDV were significantly increased from 0 to 36 h(P<0.05),and died from 36 to 48 h after infection.After infection,serum IL-8 level was increased significantly from 24 h to 60 h(P<0.05).Levels of TNF-α,TGF-β,IL-10 and MCP-1 had no significant changes without death occurrence.The serum levels of IL-8,IL-10,TGF-β and MCP-1 of adult rabbits were significantly higher than those of young rabbits after infection(P<0.05),and the level of TNF-α was higher than that of young rabbits at 24 h after infection(P<0.001),and lower than that of young rabbits at the rest of the time(P<0.01).These results indicated that the difference in serum levels of IL-8,IL-10,TNF-α,TGF-β and MCP-1 may be the key factor affecting the difference in the pathogenesis of RHDV between young and adult rabbits.
