摘要
目的 探讨lncRNA SPRR2C是否参与竹黄颗粒剂干预银屑病过程。方法 咪喹莫特(IMQ)诱导构建小鼠银屑病模型,检测不同浓度竹黄颗粒剂对SPRR2C表达的影响。IL-22处理角质形成细胞HaCaT构建银屑病细胞模型,并利用竹黄颗粒含药血清干预,其后检测SPRR2C及IL-1β、IL-6、TNF-α的表达和细胞增殖。验证SPRR2C过表达对竹黄颗粒干预的影响。结果 与对照组相比,竹黄颗粒干预组SPRR2C表达下调,干预浓度越高,SPRR2C表达水平越低,当浓度达到225 mg/mL时SPRR2C下调最显著(P<0.01)。不同浓度竹黄颗粒含药血清实验显示5%浓度的竹黄颗粒抑制HaCaT增殖效果最好(P<0.01)。IL-22处理下HaCaT增殖活性显著增强,IL-1β、IL-6、TNF-α和SPRR2C的表达均显著升高(P<0.01),而使用竹黄颗粒干预则可取得相反的效果。并且过表达SPRR2C可降低IL-22对HaCaT细胞增殖和炎症因子表达的诱导作用。结论 竹黄颗粒剂可通过抑制SPRR2C表达,进而降低角质形成细胞增殖、炎症反应,发挥银屑病治疗作用。
Objective To explore whether lncRNA SPRR2 C can be used as a biomarker to assess the intervention effect of Zhuhuang granule on psoriasis.Methods The mouse psoriasis model was induced by imiquimod under the intervention of different concentrations of Zhuhuang granule.The expression level of SPRR2 C was detected.Keratinocytes HaCaT were incubated with IL-22 to construct a psoriasis cell model and treated with Zhuhuang granule-containing serum.Cell proliferation and the expression of SPRR2 C and IL-1β,IL-6,TNF-α were analyzed.Results Compared with the control group, the expression of SPRR2 C in the Zhuhuang granule intervention group decreased.The higher the granule concentration, the lower the expression level of SPRR2 C.SPRR2 C decreased significantly when the granule concentration reached 225 mg/mL.The serum test with different concentrations of Zhuhuang granule showed that the 5% of Zhuhuang granule had the best inhibitory effect on the proliferation of HaCaT.Cell experiment showed that the proliferation activity of HaCaT was significantly enhanced and the expression of IL-1β,IL-6,TNF-α and SPRR2 C were increased significantly after treatment with IL-22.In contrast, the treatment with Zhuhuang granule-containing serum showed the opposite effect.SPRR2 C over-expression could reduce the inducing effects of IL-22 on the proliferation of HaCaT and expression of inflammatory cytokines. Conclusion Zhuhuang granule can inhibit the expression of SPRR2 C,reduce keratinocyte proliferation and inflammatory response, and then play a role in treating psoriasis.
