基于网络药理学研究苏合香冰片配伍对急性心肌梗死的作用机制 被引量:10

Action Mechanism of Styrax-Borneol Drug Pair in Treating Acute Myocardial Infarction Based on Network Pharmacology

摘要 目的:筛选“苏合香-冰片”有效成分,运用多数据库预测各有效成分及急性心肌梗死的相关靶点,构建“药物-有效成分-靶点”相互作用网络,探讨“苏合香-冰片”对急性心肌梗死的作用机制。方法:通过中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)和SwissADME数据库筛选“苏合香-冰片”的有效成分,利用Swiss Target Prediction数据库预测各有效成分的靶点,NIMNT数据库进行适应症预测,GeneCards,DisGeNET,DigSee数据库预测急性心肌梗死的共同靶点,采用网络绘图软件Cytoscape 3.7.2构建“药物-有效成分-靶点”相互作用网络,最后通过Metascape数据库和NIMNT数据库进行基因本体(Gene Ontology,GO)功能富集分析、京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路富集分析。结果:筛选得到苏合香有效成分26个,冰片有效成分13个,“苏合香-冰片”的唯一靶点共461个,其与阿尔兹海默症、冠状动脉疾病、心肌梗死、动脉硬化等有关。预测得到“苏合香-冰片”与急性心肌梗死的共同靶点共69个,主要通过参与血液循环,氧化应激等生理过程,调控磷脂酰肌醇-3激酶(Phosphatidylinositol-3-Kinases,PI3K)/蛋白激酶B(Protein Kinase B,Akt)信号通路、低氧诱导因子-1(Hypoxia-Inducible Factor-1,HIF-1)信号通路、TOLL样受体(Toll-like Receptors,TLR)信号通路及血管内皮生长因子(Vascular Endothelia Growth Factor,VEGF)信号通路等对急性心肌梗死发挥保护作用。结论:通过网络药理学研究“苏合香-冰片”对急性心肌梗死的作用机制不仅反映了中药多成分、多靶点的特点,也为后续研究“苏合香-冰片”对急性心肌梗死的作用机制提供了参考依据。 Objective:Screen the active ingredients of‘Styrax-Borneol’,use multiple databases topredict each active ingredient and acute myocardial infarction related targets,construct a‘drug-active ingredients-target’interaction network,and explore the effect mechanism of‘Styrax-Borneol’on acute myocardial infarction.Methods:Screen the active ingredients of‘Styrax-Borneol’through the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and SwissADME database,use the Swiss Target Prediction database to predict targets of each active ingredient,and the NIMNT database to predict‘Styrax-Borneol’adaptation disease.In addition,the GeneCards,the DisGeNET and DigSee databases to predict the common targets of acute myocardial infarction.The network drawing software Cytoscape 3.7.2 is used to construct the‘drug-active ingredient-target’interaction network.Finally,use the Metascape database to process gene ontology function enrichment analysis,Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis.Results:26 active ingredients of Styrax and 13 active ingredients of Borneol were screened.There are 461 unique targets of‘Styrax-Borneol’,which are related to Alzheimer’s disease,coronary artery disease,myocardial infarction,and arteriosclerosis.A total of 69 common targets of‘Styrax-Borneol’and acute myocardial infarction were predicted.They mainly participating in physiological processes such as blood circulation and oxidative stress and protecting against acute myocardial infarction through regulate phosphatidylinositol-3-kinases(PI3K)/protein kinase B(Akt)signaling pathway,Hypoxia-Inducible Factor-1(HIF-1)signaling pathway,TOLL-like receptor(TLR)signaling pathway and vascular endothelia growth factor(VEGF)signaling pathway.Conclusion:The study of the mechanism of‘Styrax-Borneol’on acute myocardial infarction through network pharmacology not only reflects the multi-component and multi-target characteristics of traditional Chinese medicine,but also provides a reference for the follow-up study of the effect mechanism of‘Styrax-Borneol’on acute myocardial infarction.
