长链非编码RNA UCA1通过调控miR-135a促进甲状腺乳头状癌的侵袭和转移

摘要 目的 研究长链非编码RNA(lncRNA)尿路上皮癌胚抗原(UCA1)在甲状腺乳头状癌中的表达及对生物学行为的影响和机制。方法 qPCR检测lncRNA UCA1和miR-135a在甲状腺癌和癌旁正常甲状腺组织以及甲状腺癌细胞株和正常甲状腺滤泡细胞株中的表达情况。双荧光酶报告基因检测lncRNA UCA1和miR-135a的相互作用。CCK-8、平板克隆实验和Transwell小室实验研究细胞生物学行为。结果 在甲状腺乳头状癌组织中lncRNA UCA1的表达明显高于癌周正常甲状腺组织。荧光素酶实验显示在乳头状癌细胞中lncRNA UCA1和miR-135a可以直接结合。lncRNA UCA1沉默后乳头状癌细胞的活性降低,克隆能力、侵袭能力和迁移能力均减弱。而在lncRNA UCA1沉默的细胞中沉默miR-135a可以逆转lncRNA UCA1沉默后对细胞生物学的影响。结论 lncRNA UCA1通过负性调控miR-135a而促进细胞的侵袭和转移。 Objective To illustrate the expression, effect on biological behavior and mechanism of Long non-coding RNA(lncRNA) urothelial carcinoma-associated 1(UCA1) in papillary thyroid cancer.Methods qPCR was used to show the expression of lncRNA UCA1 and miR-135 a in the tumor tissues and paraneoplastic normal thyroid tissue as well as thyroid cancer cell lines and normal thyroid follicular cell lines.The dual-luciferase assay was used to illustrate the correlation of lncRNA UCA1 and miR-135 a.CCK-8,colony formation assay and transwell assay was used to analyze the cell biological behavior.Results The expression of lncRNA UCA1 was significantly higher in papillary thyroid cancer tissues than in paraneoplastic normal thyroid tissue.The dual-luciferase assay showed that lncRNA UCA1 could directly connect with miR-135 a in papillary carcinoma cells.The activity of papillary carcinoma cells was reduced after lncRNA UCA1 silencing, and their clonogenic, invasive and migratory abilities were diminished, while miR-135 a silencing could reverse these effects.Conclusion lncRNA UCA1 could enhance the thyroid papillary cancer migration and invasion via negatively regulating miR-135 a.
