MMP-14、HSP27、miR-26a与结肠癌临床病理特征的关系 被引量:3

Associations of MMP-14,HSP27 and miR-26a with clinicopathological characteristics of colon cancer

摘要 目的探究基质金属蛋白酶-14(matrix metalloproteinase-14,MMP-14)、热休克蛋白27(heat shock protein 27,HSP27)、微小RNA-26a(microRNA-26a,miR-26a)在结肠癌中的表达及与临床病理特征的关系。方法选取进行根治术标本手术治疗的结肠癌患者50例,取患者手术切除癌组织、癌旁组织标本进行免疫组化染色,Western blot法检测MMP-14、HSP27表达,RT-PCR检测miR-26a表达,统计学分析通过Pearson相关性分析结肠癌组织中MMP-14、HSP27、miR-26a表达相关性。结果与癌旁组织比较,癌组织MMP-14、HSP27表达较高,miR-26a表达较低(P<0.05)。与肿瘤最大径≤3 cm患者相比,肿瘤最大径>3 cm患者MMP-14、HSP27表达较高,miR-26a表达较低(P<0.05);与Ⅰ+Ⅱ期相比,Ⅲ+Ⅳ期患者MMP-14、HSP27表达较高,miR-26a表达较低(P<0.05);与无淋巴结转移患者相比,有淋巴结转移患者MMP-14、HSP27表达较高,miR-26a表达较低(P<0.05);与高、中分化患者相比,低分化患者MMP-14、HSP27表达较高,miR-26a表达较低(P<0.05)。结肠癌组织中MMP-14、HSP27两者表达呈正相关(r=0.455,P=0.001);结肠癌组织中MMP-14、miR-26a呈负相关(r=-0.673,P=0.001);结肠癌组织中HSP27、miR-26a呈负相关(r=-0.548,P=0.001)。结论MMP-14、HSP27、miR-26a在结肠癌组织中呈现异常表达,三者表达变化与结肠癌组织大小、病理分期、淋巴结转移、组织分化等临床病理特征密切相关,并且三者在结肠癌组织中的表达之间具有相关性,表明在结肠癌发生演进过程中共同发挥重要作用。 Objective To investigate matrix metalloproteinase-14(MMP-14),heat shock protein 27(HSP27),microRNA-26a(miR-26a)in colorectal adenocarcinoma and their correlations with the clinicopathological features.Methods A total of 50 colon cancer patients who underwent radical resection treatment were selected.Tumor tissue samples and paracancer tissue samples were collected from the patients for immunohistochemical staining.Western blot was used to assess the expression of MMP-14 and HSP27;and RT-PCR was used to assess the expression of miR-26a.The correlations analysis was performed on the expression of MMP-14,HSP27 and miR-26a in colon cancer tissues by Pearson correlation.Results Compared with para-cancer tissues,the expressions of MMP-14 and HSP27 were significantly higher in cancer tissues,while the expression of miR-26a was significantly lower(P<0.05).Compared with patients with tumor diameter≤3 cm,patients with tumor diameter>3 cm had significantly higher expressions of MMP-14 and HSP27,and significantly lower expression of miR-26a(P<0.05).Compared with StageⅠ+Ⅱ,the expressions of MMP-14 and HSP27 were significantly higher in stageⅢ+Ⅳ,and the expression of miR-26a was significantly lower(P<0.05).Compared with patients without lymph node metastasis,the expressions of MMP-14 and HSP27 were significantly higher in patients with lymph node metastasis,and the expression of miR-26a was significantly lower in patients with lymph node metastasis(P<0.05).Compared with the patients with high and medium differentiation,the expressions of MMP-14 and HSP27 were significantly higher and the expression of miR-26a was significantly lower in the patients with low differentiation(P<0.05).There was a positive correlation between MMP-14 and HSP27 expression in adenocarcinoma tissues(r=0.455,P=0.001).There was a negative correlation between MMP-14 and miR-26a in colon cancer tissues(r=-0.673,P=0.001).There was a negative correlation between HSP27 and miR-26a in colon cancer tissues(r=-0.548,P=0.001)Conclusion MMP-14,HSP27 and miR-26 a present abnormal expression in the colon cancer tissue.They are correlated with the size,pathologic stage,lymph node metastasis and differentiation of colon cancer,suggesting important roles in colon cancer.
