基于免疫调节作用探讨肺康养阴颗粒对于肺系疾病恢复期的干预作用

Effect of Feikang Yangyin Granule on Pulmonary Disease in Convalescence Based on Immune Regulation

目的探讨肺康养阴颗粒对肺系疾病恢复期的干预作用及其可能的作用机制。方法通过考察肺康养阴颗粒对脂多糖(LPS)所致大鼠急性肺损伤的保护作用和对实验性免疫功能低下小鼠的免疫调节作用,研究肺康养阴颗粒对肺疾病的干预作用及其可能机制。结果对LPS所致大鼠急性肺损伤的实验中,肺康养阴颗粒高剂量组(10.8 g生药/kg)大鼠全肺和肺系数明显降低(P<0.05),脾和胸腺重量以及脾和胸腺系数均显著增加(P<0.05,P<0.01);肺康养阴颗粒低、中、高剂量组的左肺湿重均不同程度下降(P<0.05,P<0.01),高剂量组的肺组织湿/干比(W/D)也同样明显下降(P<0.05);肺康养阴颗粒的高剂量组TNF-α显著降低(P<0.01);肺康养阴颗粒的各剂量组血清中IL-6含量较模型组有降低趋势。在环磷酰胺制备的小鼠免疫功能低下模型实验中,发现肺康养阴颗粒低剂量组(11.31 g生药/kg)、高剂量组(45.24 g生药/kg)能明显提高环磷酰胺所致免疫功能低下小鼠网状内皮系统吞噬能力,表明本品具有一定的增强非特异性免疫功能的作用;肺康养阴颗粒各剂量组可明显增强环磷酰胺所致免疫功能低下小鼠血清溶血素生成的作用,表明本品具有一定的增强体液免疫功能的作用。结论肺康养阴颗粒能显著减轻LPS诱导急性肺损伤大鼠的肺水肿程度,显著降低大鼠血清中的炎性细胞因子含量,对LPS诱导急性肺损伤大鼠具有较好的保护作用,抗炎效果明显,其可能的机制为提高机体免疫功能,降低炎症因子含量而发挥抗炎作用。以上结果为临床应用肺康养阴颗粒干预肺疾病提供了药效学依据。

Objective To explore the intervention effect and possible mechanism of Feikang Yangyin Granule on the convalescent stage of pulmonary diseases.Methods To investigate the protective effect of Feikang Yangyin Granule on lipopolysaccharide(LPS)-induced acute lung injury in rats and its immunomodulatory effect on experimental immunosuppressed mice and study the intervention effect of Feikang Yangyin Granule on lung disease and its possible mechanism.Results In the experiment of acute lung injury induced by LPS,the whole lung and lung coefficients of rats were obviously decreased(P<0.05).The weights of spleen and thymus gland and the coefficient of spleen and thymus gland were increased significantly(P<0.05,P<0.01)in the high dose group of Feikang Yangyin Granule(10.8 g crude drug/kg).The wet weights of left lung were obviously decreased(P<0.05,P<0.01)in the low,middle and high dose groups of Feikang Yangyin Granule in different degrees,and the wet/dry ratio(w/d)of lung tissue was also decreased(P<0.05).The content of TNF-αin high dose group of Feikang Yangyin Granule was decreased significantly(P<0.01).The content of IL-6 in serum of each dose group of Feikang Yangyin Granule was lower than that of the model group.In the mouse model of immunosuppression prepared by the cyclophosphamide,it was found that the phagocytosis of reticuloendothelial system was significantly improved in the low(11.31 g crude drug/kg)and high(45.24 g crude drug/kg)dose groups of Feikang Yangyin Granule,indicating that this product had the function of enhancing the Hapten function.Each dose group of Feikang Yangyin Granule could obviously enhance the effect of serum hemolysin production in immunosuppressed mice induced by cyclophosphamide,indicating that this product had the function of enhancing humoral im-munity.Conclusion Feikang Yangyin Granule can significantly reduce the degree of pulmonary edema and the contents of inflammatory cytokines in the serum of rats with LPS-induced acute lung injury.It has a good protective effect on LPS-induced acute lung injury in rats and has obvious anti-inflammatory effect.Its possible mechanism is to improve the body's immune function,reduce the contents of inflammatory factors and play an anti-inflammatory role.These results provide the pharmacodynamic basis for clinical application of Feikang Yangyin Granule to intervene lung disease.
