摘要
目的检测糖尿病模型鼠视网膜中GALNT2、P-EGFR的表达,并与对照组比较,为进一步研究GALNT2介导EGFR对糖尿病视网膜病变的作用机制奠定基础。方法将雄性Wistar大鼠分为模型组6例:腹腔注射2%STZ溶液(50 mg/kg)以诱发STZ糖尿病;对照组6例:注射同等体积的柠檬酸-柠檬酸钠缓冲液。实时定量PCR检测两组视网膜中GALNT2 mRNA;免疫印迹法检测GALNT2、P-EGFR的表达。结果实时定量PCR检测:与对照组相比,实验组糖尿病模型鼠视网膜中GALNT2 mRNA的表达显著下调,差异有统计学意义(P<0.01);免疫印迹法检测:与对照组相比,实验组糖尿病模型鼠视网膜中GALNT2蛋白的表达显著下调,差异有统计学意义(P<0.01);P-EGFR蛋白表达显著升高,差异有统计学意义(P<0.01)。结论糖尿病模型鼠视网膜中GALNT2的下调,促进了EGFR的磷酸化,从而对糖尿病视网膜病变起到了保护作用。
Objective To detect the expression of GALNT2 and P-EGFR in the retina of diabetic mice,and to compare the expression with that of the control group,so as to lay a foundation for the further study of the mechanism of GALNT2 mediated EGFR on diabetic retinopathy.Methods Male Wistar rats were divided into model group(n=6),which was intraperitoneally injected with 2%STZ solution(50 mg/kg)to induce STZ diabetes,and control group(n=6),which was injected with citric acid-sodium citrate buffer of the same volume.Real-time quantitative PCR was used to detect GALNT2 mRNA in retina of both groups.Western blotting was used to detect the expression of GALNT2 and P-EGFR.Results Real-time quantitative PCR detection:compared with the control group,the expression of GALNT2 mRNA in the retina of diabetic model rats in experimental group was significantly down-regulated,and the difference was statistically significant(P<0.01).Western blotting:compared with the control group,the expression of GALNT2 protein in the retina of diabetic rats in experimental group was significantly down-regulated(P<0.01).The expression of P-EGFR protein was significantly increased,and the difference was statistically significant(P<0.01).Conclusion The down-regulation of GALNT2 in the retinas of diabetic mice promoted the phosphorylation of EGFR,thus playing a protective role in diabetic retinopathy.The down-regulation of GALNT2 in the retinas of diabetic mice promoted the phosphorylation of EGFR,thus playing a protective role in diabetic retinopathy.
