地参总黄酮的提取及抗氧化活性研究

摘要 目的:研究地参总黄酮的超声提取工艺及体外抗氧化活性。方法:以地参为原料,乙醇为提取剂,运用超声波辅助法从地参中提取总黄酮,在单因素实验基础上用正交实验优化提取工艺。采用DPPH·法对地参总黄酮的自由基清除能力和总抗氧化活性进行评价。结果:地参总黄酮最佳提取工艺条件为:乙醇体积分数为70%,超声功率为175 W,超声时间为20 min,料液比为1∶30。总黄酮提取率高达96.25 mg·g^(-1)。超声提取法得到的地参提取物对DPPH·有很好的清除作用,IC_(50)值为0.00272 mg·mL^(-1)。结论:优选的总黄酮提取工艺可行,地参总黄酮有较好的抗氧化作用,为其研究开发提供一定的参考依据。 The ultrasonic extraction technology and in vitro antioxidant activity of total flavonoids in Lycopus lucidus Turcz.were studied.Ethanol was used as the extractant to extract the total flavonoids in Lycopus lucidus by ultrasonic-assisted extraction, and the extraction process was optimized through single factor tests and orthogonal experiment.The DPPH· free radical scavenging capacity and the total antioxidant activity of the total flavonoids were evaluated.The optimum extraction process was as follows: the ethanol concentration was 70 %,the extraction power was 175 W,the extraction time was 20 min, and the solid-liquid ratio was 1∶30.The extracted total flavonoids content reached 96.25 mg/g.The total flavonoids had good scavenging effect on DPPH· free radical, with an IC_(50) value of 0.00272 mg/mL.The ultrasonic extraction of total flavonoids in Lvcopus lucidus is feasible, and the extracts by this method have high antioxidant activity.The study has provided reference for the research and development of Lycopus lucidus.
