血人参提取物及其制剂对半乳糖联合亚硝酸钠致衰老小鼠模型抗氧化能力的影响 被引量:1

摘要 目的:研究血人参提取物及其制剂对半乳糖联合亚硝酸钠致衰老小鼠模型抗氧化能力的影响,探讨其作用机制。方法:将雄性KM小鼠进行水迷宫实验,随机分为阳性对照VitE(0.75 g·kg^(-1))组,芪胶升白胶囊高、中、低剂量(2 g·kg^(-1)、1 g·kg^(-1)、0.5 g·kg^(-1))组,血人参乙酸乙酯部位高、中、低剂量(0.33 g·kg^(-1)、0.17 g·kg^(-1)、0.08 g·kg^(-1))组,每组20只,除正常组与模型组(以皮下注射150 mg·kg^(-1) D-半乳糖联合腹腔注射50 mg·kg^(-1)亚硝酸钠构建小鼠衰老模型)外,其余各组造模给药同时进行,连续8周。每周测体重,第7周进行水迷宫实验,第8周取脏器计算脏器指数;测定血清以及脑组织、肝脏、肾脏和心脏匀浆上清液中超氧化物歧化酶(SOD)活性、谷胱甘肽过样化物酶(GSH-Px)和丙二醛(MDA)含量。结果:与模型组比较,各给药组均能增加衰老小鼠体质量,对脏器指数无明显影响,提高学习记忆能力,血人参乙酸乙酯部位与芪胶升白胶囊各剂量组均能提高血清、脑及各脏器组织中的SOD与GSH-Px活性,降低部分组织中MDA含量,以高剂量组效果最显著。结论:血人参提取物及其制剂能增加机体SOD和GSH-Px活力,降低MDA含量,抑制脂质过氧化物的形成而起到延缓衰老的作用。 We studied the effect of Indigofera staclrvoides extract and its preparation on the antioxidant capacity in aging mice model induced by galactose combined with sodium nitrite, and its mechanism of action was explored.Male KM mice were subjected to water maze experiment, and randomly divided into positive control VitE group(0.75 g/kg),Qijiao Shengbai capsule high-,medium-and low-dose group(2 g/kg, 1 g/kg, 0.5 g/kg),Indigofera staclrvoides ethyl acetate part high-,medium-and low-dose group(0.33 g/kg, 0.17 g/kg, 0.08 g/kg).Each group contained 20 mice.Except for the normal group and the model group(subcutaneous injection of 150 mg/kg D-galactose combined with intraperitoneal injection of 50 mg/kg sodium nitrite to construct the aging model),the other groups were administered simultaneously for 8 consecutive weeks.The body weight of the mice was measured every week, the water maze experiment was carried out in the 7 th week, and the organs are taken to calculate the organ indexes in the 8 th week.The superoxide dismutase(SOD)activity in the serum and the supernatant of brain tissue, liver, kidney and heart was measured, as well as the contents of glutathione cross-like enzyme(GSH-Px)and malondialdehyde(MDA).Compared with the model group, the body weight of the aging mice in each administered group increased without significant effect on the organ indexes, and their learning and memory ability was improved.Both the ethyl acetate part of Indigofera staclrvoides and the Qijiao Shengbai capsule could increase the activity of SOD and GSH-Px in serum, brain and various organs and tissues, while reducing the content of MDA in some tissues.The high-dose group had the most significant effect.In conclusion, Indigofera staclrvoides extract and its preparation can increase the activity of SOD and GSH-Px in the body, reduce the content of MDA,and inhibit the formation of lipid peroxides, thereby having the effect of delaying aging.
