乌江渡水库沉积物风干和过筛处理与原始沉积物中微生物群落的差异比较分析

摘要 本研究对乌江渡水库的沉积物细菌群落进行风干和过筛处理,旨在比较不同处理后沉积物与原始细菌群落结构的差异,从而为以后此类样品的处理提供理论依据。结果显示乌江渡水库沉积物中细菌的优势类群主要为变形菌门(Proteobacteria),包括γ-Proteobacteria,Δ-Proteobacteria和α-Proteobacteria。其群落结构在风干和过筛处理后有一定程度的改变,表现为优势类群相对丰度的变化。此外,多样性指数和基因功能差异不显著。然而风干和过筛处理之间群落结构无显著差异。因此推测:风干和过筛处理中沉积物失水的过程是造成其中细菌群落结构变化的主要因素。本研究建议对水体沉积物微生物的研究应该直接使用原始保存的样品进行DNA提取,从而保存沉积物中原始微生物群落的特征。 In this study the sediment from Wujiangdu Reservoir was air-dried/filtrated, and the bacterial communities in the air-dried/filtered sediment were compared with that in the original sediment, so as to provide reference for future sample treatment.The results showed that the dominant bacterial groups in the sediment were Proteobacterial, including γ-Proteobacterial, Δ-Proteobacterial, and α-Proteobacterial.Compared with the original sediment, the bacterial community structure changed to a certain extent after air drying/filtration, with changes in the relative abundance of the dominant groups.The diversity indexes and gene functions, however, did not change significantly.There was no significant difference between air drying and filtration treatment.Therefore, the dehydration process during air drying/filtration treatment was the main reason for the change of bacterial community structure in the sediment.It is suggested that the sediment should be preserved in-situ for DNA extraction, so that the original features of the microbial communities in the sediment can be preserved.
