消癌平注射液促进肺腺癌细胞凋亡的分子机制研究 被引量:2

摘要 目的:研究消癌平注射液对A549、H1299细胞增殖的影响,并初步探讨其相关的分子机制。方法:体外培养肺癌细胞A549、H1299,用不同浓度的消癌平注射液处理48 h。采用MTT法检测消癌平注射液的增殖抑制效率;采用流式细胞术检测各组细胞凋亡及细胞周期抑制情况;采用Western blot检测GSK-3β、NF-κB、Bax、Bcl-2等蛋白的表达量。结果:A549、H1299细胞经消癌平注射液处理后,与对照组相比,细胞的增殖抑制率以剂量依赖性方式增加(P<0.05);不同浓度消癌平注射液作用后,与对照组相比较,细胞凋亡率增加;不同浓度消癌平注射液处理后,停滞于G0/G1期细胞增多。Western blot显示NF-κB、p-GSK-3β^(Ser9)、Bcl-2蛋白表达量随着消癌平注射液浓度的增加而降低。结论:消癌平注射液可促进肺腺癌细胞A549、H1299的凋亡,其机制可能是通过调控GSK-3β-NF-κB-Bcl-2轴发挥作用。 Objective:To investigate the effect of Xiaoaiping injection on and its related molecular mechanism in proliferation of A549 and H1299 cells.Methods:Human alveolar cells A549 and H1299 were cultured in vitro,and then treated with different dose of Xiaoaiping injection for 48 h.MTT assay was perfomred to detect the proliferation inhibition efficiency of Xiaoaiping injection.Flow cytometry was used to detect the cell apoptosis and cell cycle inhibition in each group.Western blot was done to measure the protein expression levels of GSK-3β,NFκB,Bax and Bcl-2.Results:Compared with the control group,inhibition rate of cell A549 and H1299 proliferation was increased in a dose-dependent manner,and apoptosis rate of the cells was also increased after treatment with Xiaoaiping injection(P<0.05),particularly the cells were increased at G0/G1 phase following Xiaoaiping injection treatment in different dose.Western blot assay revealed that the protein expression of NF-κB,p-GSK-3β^(Ser9) and Bcl-2 was decreased with added dose of Xiaoaiping injection.Conclusion:Xiaoaiping injection can induce the apoptosis of lung adenocarcinoma cell A549 and H1299.The potential mechanism may be involved in the axis of GSK-3β-NF-κB-Bcl-2 being regulated.
