种猪常温精液新旧标准检测方法的比较研究 被引量:4

摘要 【目的】为了研究基于CASA系统等新检测方法对种猪常温精液品质检测的影响,选取36份种猪常温精液样品开展品质检测与对比分析。【方法】剂量检测采用称量法与量筒法;精子活力、前向运动精子数检测采用人工目测评分法与CASA系统检测法;精子畸形率检测采用5种快速染色方法。【结果】剂量检测称量法与量筒法比较,精子活力检测不同CASA系统比较,以及CASA系统与质检中心检验员比较,前向运动精子数检测CASA系统与血球计数板法比较,差异均不显著(P>0.05);精子活力检测CASA系统与不同企业检验员比较差异显著(P<0.05);精子畸形率姬姆萨染色法缩短染色时长至15~30min染色效果良好,筛选5种染色方法后确定伊红苯胺黑染色法为快速检测方法。【结论】剂量检测称量法优于量筒法,且不会对精子产生损害;精子活力和前向运动精子数检测采用CASA系统更加准确、客观、高效;精子畸形率检测采用改进姬姆萨染色法和伊红苯胺黑快速染色法,均可达到快速、准确的目的。新方法在行业推广应用中具有很好的应用价值。 [Objective] The quality of 36 semen samples were analyzed with different detection methods based on the CASA system, to study the influence of new detection methods on the quality detection of boar liquid semen. [Method] The weighing method and the measuring cylinder method were selected for dose detection;sperm motility and the number of progressively motile sperm were detected by artificial visual scoring and the CASA system;sperm deformity rate was determined by 5 rapid staining methods. [Result] There was no significant difference between the weighing method and the measuring cylinder method for dose detection(P>0.05), no significant difference between CASA systems for sperm motility detection(P>0.05), and between the CASA system and examiner(P>0.05), no significant difference between CASA systems and the hemocytometer method for the number of progressively motile sperm(P>0.05). However, there was significant difference between the CASA system and the personnel of different enterprises for sperm motility detection(P<0.05);the sperm deformity rate of giemsa staining method shortens the staining time to 15min~30min;the eosin aniline black staining method was selected as the rapid detection method through the screening of 5 rapid sperm staining methods. [Conclusion] The dosage weighing method is better than the measuring cylinder method, which does not cause damage to sperm;it is more accurate, objective and efficient to detect sperm motility and the number of progressively motile sperm than using the CASA system;it is more rapid and accurate to detect sperm deformity rate with the short time giemsa staining method and eosin aniline black staining method.
