蒙汉民族ASAP1、NRAMP1、SP110基因表达水平与结核病易感性关联分析 被引量:1

Correlation Between Expression Levels of ASAP1,NRAMP1,SP110 Genes and Susceptibility to Tuberculosis in Mongolian and Han Nationalities

摘要 结核病(TB)发病具有种族易感性的分布特点,揭示结核病种族易感性的发病机制对明确结核病的高危人群、制定相应的防控策略具有重要作用。本研究从宿主易感相关基因转录水平出发,探讨ASAP1、NRAMP1和SP110表达水平与蒙古族和汉族结核病发病的关联程度。基于病例-对照研究设计,收集蒙古族和汉族96例结核病患者和126例健康人群静脉血,采用实时荧光定量PCR的方法检测易感性关联基因ASAP1、NRAMP1和SP110的mRNA表达水平,并分析不同民族间、结核患者与健康对照组间基因表达水平的差异。蒙古族结核患者结核易感基因ASAP1、NRAMP1 mRNA表达水平明显高于对照组(P<0.05),汉族结核病患者NRAMP1表达水平高于对照组(P<0.05)。蒙古族女性结核病患者,SP110的表达显著性差异(P<0.05)。汉族ASAP1、SP110 mRNA表达在结核病患者与对照之间没有显著差异。蒙汉民族结核病发病易感性与宿主基因ASAP1、NRAMP1和SP110表达有关,尤其是女性人群。提示结核病防治过程中可参考这些易感基因的表达评估结核病高危人群。 Tuberculosis(TB)has the susceptibility trending among ethnic groups.In order to investigate if racial predisposition plays an important role in the TB pathogenesis among the high-risk groups and carry out the corresponding prevention and control strategies,the present study explored the correlation of the expression levels of ASAP1,NRAMP1 and SP110 genes between Mongolian and Han TB patients.Based on the case-control study design,venous blood samples of 96 Mongolian and Han TB patients and 126 healthy people were collected for extraction of total RNA templates from which cDNA materials were generated.Then mRNA expression levels of ASAP1,NRAMP1 and SP110 genes were quantified by real-time fluorescence quantitative PCR.The results showed that the mRNA expression levels of TB susceptibility associated ASAP1 and NRAMP1 genes in Mongolian patients was significantly higher(P<0.05)than those of healthy Mongolian control group while significant difference(P<0.05)in the mRNA expression level of NRAMP1 gene existed between Han TB patients and healthy control group.Moreover,the significant difference(P<0.05)in the mRNA expression level of SP110 gene was found in female Mongolian TN patients but no difference in the mRNA expression levels of ASAP1 and NRAMP1 genes was found between female Han TB patients and healthy controls.In summary,TB susceptibility correlated with the expression levels of ASAP1,NRAMP1 and SP110 genes in Mongolian and Han ethnic groups particularly the female populations,suggesting that the expression of these TB susceptibility associated genes could be referred to an indicator of high-risk groups,giving information for implementation of effective preventive measures for TB high risk populations.
