藏红花素对慢性阻塞性肺疾病急性加重期大鼠 肺组织NOD样受体蛋白3表达及血清IL-1β水平的影响 被引量:8

摘要 目的:探讨藏红花素对慢性阻塞性肺疾病(COPD)急性加重期大鼠NOD样受体蛋白3(NLRP3)表达及血清白细胞介素-1β(IL-1β)水平的影响。方法:将40只大鼠随机分为对照组、模型组和藏红花素小、大剂量组。采用肺功能检测仪检测大鼠肺功能指标,TUNEL染色检测肺上皮细胞凋亡情况,比色法检测血清丙二醛(MDA)和乳酸脱氢酶(LDH)水平,RT-qPCR和Western blot法检测NLRP3表达,ELISA法检测大鼠血清IL-1β水平。结果:与对照组比较,模型组大鼠呼气峰流量(PEF)、吸气峰流量(PIF)、0.3秒用力呼气容积与用力肺活量比值(FEV0.3/FVC)显著降低(均P<0.05),肺泡上皮细胞凋亡率、肺组织NLRP3表达水平以及血清IL-1β、MDA和LDH水平显著升高(均P<0.05)。与模型组比较,藏红花素小、大剂量组大鼠PEF、PIF、FEV0.3/FVC显著升高(均P<0.05),肺泡上皮细胞凋亡率、肺组织NLRP3表达水平及血清IL-1β、MDA和LDH水平显著降低(均P<0.05)。结论:藏红花素可有效改善COPD急性加重期大鼠的肺功能,抑制氧化应激和炎症反应,减少肺泡上皮细胞凋亡,其机制与下调NLRP3表达有关。 Objective:To investigate the effect of crocin on the expression of NLRP3 and the level of serum interleukin(IL)-1βin chronic obstructive pulmonary disease(COPD)acute exacerbation rats.Methods:Forty rats were randomly divided into control group,model group,low and high dose groups of crocus.Pulmonary function tester was used to detect relevant pulmonary function indexes in rats,TUNEL staining was used to detect the apoptosis of lung epithelial cells,the levels of malondialdehyde(MDA)and lactate dehydrogenase(LDH)in serum were detected by colorimetry,RT-qPCR and Western blot were applied to detect NOD-like receptor protein 3(NLRP3)expression,and ELISA was used to detect the level of IL-1βin serum.Results:Compared with the control group,the related pulmonary function indexes including peak expiratory flow(PEF),peak inspiratory flow(PIF),and the ratio of forced expiratory volume in 0.3 seconds to forced vital capacity(FEV0.3/FVC)of rats in the model group were significantly decreased(all P<0.05),the apoptosis rate of alveolar epithelial cells,the expression level of NLRP3 in lung tissue,and the level IL-1β,MDA and LDH in serum were significantly increased(all P<0.05).Compared with the model group,the related pulmonary function indexes PEF,PIF,FEV0.3/FVC in the low and high dose groups of crocus were significantly increased(all P<0.05),and the apoptosis rate of alveolar epithelial cells,the expression level of NLRP3 in lung tissue,and the level of IL-1β,MDA and LDH in serum were significantly decreased(all P<0.05).Conclusion:Crocin could effectively improve lung function in rats with acute exacerbation of COPD,inhibit oxidative stress and inflammatory response,and reduce alveolar epithelial cell apoptosis,and its mechanism was related to down-regulation of NLRP3 expression.
