摘要
目的 探讨2型糖尿病患者血清长链非编码RNA生长停滞特异性转录本5(lncRNA GAS5)与心血管疾病(CVD)风险的相关性。方法 选取2017年8月至2019年6月河北中石油中心医院收治的2型糖尿病患者248例,根据随访2年期间是否发生CVD将患者分为CVD组106例和非CVD组142例。血清lncRNA GAS5与一般资料及实验室指标相关性采用Pearson法及Spearman法分析;血清lncRNA GAS5水平对2型糖尿病患者CVD的预测价值采用受试者工作特征曲线(ROC曲线)分析;影响2型糖尿病患者发生CVD的因素采用单因素及多因素Logistic回归分析。结果 CVD组体质量指数(BMI)、低密度脂蛋白胆固醇(LDL-C)高于非CVD组,血清lncRNA GAS5低于非CVD组,差异有统计学意义(t=4.727、8.133、12.360,P<0.05)。2型糖尿病并发CVD患者血清lncRNA GAS5与BMI、舒张压、LDL-C、甘油三酯(TG)呈负相关(r=-0.524、-0.508、-0.496、-0.486,P<0.05)。ROC曲线分析结果显示,血清lncRNA GAS5预测2型糖尿病患者CVD的曲线下面积(AUC)为0.844(95%CI:0.795~0.892),特异性为0.82%,敏感度为0.75%。多因素Logistic回归分析结果显示,BMI是2型糖尿病患者发生CVD的独立危险因素(P<0.05),lncRNA GAS5是2型糖尿病患者发生CVD的保护因素(P<0.05)。结论 血清lncRNA GAS5与2型糖尿病患者CVD的发生有关,可能作为2型糖尿病CVD风险的预测指标。
Objective To investigate the correlation between serum long non-coding RNA growth arrest specific transcript 5(lncRNA GAS5)and the risk of cardiovascular disease(CVD)in patients with type 2diabetes. Methods A total of 248 patients with type 2 diabetes admitted to Hebei PetroChina Central Hospital from August 2017 to June 2019 were selected,the patients were divided into CVD group with 106 cases and nonCVD group with 142 cases. the correlation between serum lncRNA GAS5 and general data and laboratory indicators was analyzed by Pearson method and Spearman method;the predictive value of serum lncRNA GAS5levels for CVD in patients with type 2 diabetes was analyzed by receiver operating characteristic curve(ROC curve);the factors affecting the occurrence of CVD in patients with type 2 diabetes were analyzed by single factor and multivariate Logistic regression analysis. Results The body mass index(BMI)and low-density lipoprotein cholesterol(LDL-C)of the CVD group were higher than those of the non-CVD group,and the serum lncRNA GAS5 was lower than that of the non-CVD group,the difference was statistically significant(t=4.727,8.133,12.360,P<0.05). Serum lncRNA GAS5 of patients with type 2 diabetes complicated with CVD was negatively correlated with BMI,diastolic blood pressure,LDL-C,and triglycerides(TG)(r=-0.524,-0.508,-0.496,-0.486,P<0.05). ROC curve analysis results showed that the area under the curve(AUC)of serum lncRNA GAS5 predicting CVD in patients with type 2 diabetes was 0.844(95%CI:0.795~0.892),the specificity was 0.82%,and the sensitivity was 0.75%. The results of multivariate Logistic regression analysis showed that BMI was an independent risk factor for CVD in patients with type 2 diabetes(P<0.05),and lncRNA GAS5 was a protective factor for CVD in patients with type 2 diabetes(P<0.05). Conclusion Serum lncRNA GAS5 is related to the occurrence of CVD in patients with type 2 diabetes,and may be used as a predictor of CVD risk in type 2 diabetes.
