绿茶中表没食子儿茶素没食子酸酯对炎性肠病小鼠的肠道保护作用

摘要 目的分析绿茶中表没食子儿茶素没食子酸酯(EGCG)对炎性肠病小鼠的肠道保护作用,为探究儿童炎性肠病的药学防治提供依据。方法选取SFP级6周龄、体重16-18 g的C57BL/6J小鼠15只,随机分为正常对照组、3%硫酸葡聚糖钠(DSS)组和DSS+ECCG组,每组5只。DSS组和DSS+EGCG组在饮用水中加入3%DSS,DSS+EGCG组每日灌胃给予EGCG(50mg/kg in PBS);对照组小鼠不做特殊处理。实验第7天处死所有小鼠,并取其结肠末端进行组织苏木精-伊红(HE)染色评价其病理损伤,同时将其结肠组织匀浆液进行ELISA检测,得到其中炎症因子白细胞介素6(IL-6)以及肿瘤坏死因子α(TNF-α)的变化。结果DSS造成小鼠出现腹泻、便血、体重减轻等类IBD现象,同时出现结肠长度缩短、腺体丢失、IL-6以及TNF-α增加;EGCG给药可显著缓解小鼠的组织病理学损伤,同时降低结肠组织中IL-6以及TNF-α。结论EGCG对小鼠肠道有显著的抗炎作用,可有效预防小鼠IBD的炎性损伤,是绿茶中具有肠道保护作用的关键活性物质。 Objective To analyze the intestinal protective effect of epigallocatechin gallate(EGCG)in green tea on inflammatory bowel disease mice,so as to provide basis for pharmaceutical prevention and treatment of inflammatory bowel disease in children.Method Fifeen SFP grade C57BL/6J mice aged 6 weeks and weighing 16-18 g were randomly divided into normal control group,3%dextran sulfate sodium(DSS)group and DSS+EGCG group,with 5 mice in each group.DSS group and DSS+EGCG group were added with 3%DSS in drinking water.DSS+EGCG group was given EGCG(50 mg/kg in PBS)by ga-vage every day;The mice in the control group were not given special treatment.On the 7th day of the experiment,all mice were killed,and the end of their colon was stained with hematoxylin eosin(he)to evalu-ate their pathological damage.At the same time,the colon tissue homogenate was detected by ELISA to obtain the inflammatory factor interleukin-6(IL-6)and tumor necrosis factor a(TNF-α)Changes in.Results DSS treatment led to diarhea,hematochezia,body weight reduction and other clinical IBD-like symptoms in mice,it also caused colon length shortening,gland loss,IL-6 and TNF-αaccumulation.EGCG was able to significantly alleviate pathological injury,decrease the concentrations of IL-6 and TNF-αin mice.Conclusion EGCG has significant anti-inflammatory effect on the colon tissues of mice,it can prevent inflammatory injury of IBD effectively.EGCG can serve as a key bio-active compound in green tea with intestinal protective capability.
