Bcl-2家族在脐带间充质干细胞外泌体改善阿霉素心肌损伤中的作用

摘要 目的 探讨Bcl-2家族在脐带间充质干细胞外泌体(Exo)改善阿霉素(Dox)诱导心肌损伤中潜在机制。方法 大鼠心肌细胞H9C2分为空白对照组、Dox组(0.2 mg/L Dox)、H-Exo+Dox组(30 mg/L Exo、0.2 mg/L Dox)、L-Exo+Dox组(15 mg/L Exo、0.2 mg/L Dox)。流式细胞术检测细胞凋亡,Western blot检测Bax、Bcl-2、Caspase-3表达。结果 H-Exo+Dox、L-Exo+Dox组细胞凋亡率及Bax、Caspase-3表达明显低于Dox组,而Bcl-2表达、Bcl-2/Bax比值明显增加,特别是H-Exo+Dox组(P<0.05)。结论 脐带间充质干细胞Exo通过下调Bax、Caspase-3表达和上调Bcl-2表达来减轻Dox对心肌损害。 Objective To investigate the potential mechanism of Bcl-2 family on doxorubicin(Dox)-induced myocardial injury improved by umbilical cord mesenchymal stem cell Exosomes(UCMSC-Exo). Methods Rat myocardial H9C2 cells were divided into blank control, 0.2 mg/L Dox, H-EXO +Dox(30 mg/L Exo + 0.2 mg/L Dox), and L-Exo+Dox(15 mg/L Exo + 0.2 mg/L Dox) groups. Apoptosis and expression of Bax, Bcl-2, and Caspase-3 were detected by flow cytometry and Western blot, respectively. Results Compared with Dox group, apoptosis and expression of Bax and Caspase-3 were lower, while Bcl-2 expression and Bcl-2/Bax ratio were higher in H-Exo+Dox and L-Exo+Dox groups, especially in H-Exo+Dox group(P<0.05).Conclusion UCMSC-Exo can attenuate Dox-induced myocardial injury via down-regulating Bax and caspase-3 expression and up-regulating Bcl-2 expression.
