摘要
目的观察莫西沙星治疗豚鼠实验感染鼠疫的疗效。方法参照《中华人民共和国药典》成人剂量,用动物折算系数法换算成豚鼠每日用药剂量,莫西沙星治疗组10只豚鼠,对照组10只豚鼠。鼠鼷部皮下注射鼠疫菌141株1×10;cfu/ml,于感染后24 h开始皮下注射莫西沙星治疗。实验组动物连续用药9 d,停药后观察9 d,对照组每日进行皮下注射生理盐水并观察,不进行治疗。结果莫西沙星治疗组动物经治疗后全部存活,治愈率为100%,治疗组动物在停药后继续医学观察9 d处死解剖,鼠疫细菌学培养阴性;对照组动物分别于感染后3~7 d死亡,死亡率为100%,解剖后各脏器均分离到鼠疫菌,为特异性死亡。结论莫西沙星可用于鼠疫的治疗和预防。
Objective To observe the efficacy of moxifloxacin in experiments for guinea pigs infected with plague.Methods The animal conversion coefficient method was used to convert to daily dosage of guinea pigs based on the Pharmacopoeia of The People′s Republic of China,10 guinea pigs in the treatment group was treated with moxifloxacin,and another 10 guinea pigs were admitted in the control group.A total of 141 Yersinia pestis strains 1×10~7 cfu/ml were injected subcutaneously into the sites of guinea pigs,and the treatment of moxifloxacin was initiated in 24 h after infection.The animals in the experimental group were treated with moxifloxacin for 9 days and observed for 9 days after withdrawal of medication.The guinea pigs in the control group were subcutaneously injected with normal saline dailyt and were observed without treatment.Results All the animals in treatment group treated with moxifloxacin survived after treatment,and the cure rate was 100%.The animals in the treatment group were sacrificed and dissected after continuous medical observation for 9 days,and the plague bacteriology culture was found to be negative.The animals in the control group died with the mortality rate of 100%within 3-7 days after infection.Yersinia pestis was isolated from all organs after dissection,indicating a specific death.Conclusion Moxifloxacin is eligible to be used for the treatment and prevention of plague.
