蒺藜皂苷对膀胱癌细胞增殖和凋亡的影响 被引量:3

Effect of Tribulus terrestris saponins on proliferation and apoptosis of bladder cancer cells

摘要 目的研究蒺藜皂苷调控lncRNA NKILA对膀胱癌细胞增殖、凋亡的影响及机制研究。方法将膀胱癌细胞J82分为对照组(Control组)、GSTT组、GSTT+si-NC组及GSTT+si-NKILA组。对照组用0 mg·L^(-1)蒺藜皂苷处理,GSTT组用80 mg·L^(-1)蒺藜皂苷处理,将siRNA control和NKILA siRNA转染到细胞内,细胞转染后以含有80 mg·L^(-1)蒺藜皂苷细胞培养液培养细胞,为GSTT+si-NC组及GSTT+si-NKILA组。通过细胞计数法-8(CCK-8)检测J82细胞活力;以实时荧光定量逆转录聚合酶链反应(RT-qPCR)检测NKILA的表达水平;以克隆形成实验检测各组细胞增殖能力;以流式细胞术检测J82细胞凋亡情况;以蛋白质印迹法检测凋亡蛋白水平。结果Control组、GSTT组、GSTT+si-NC和GSTT+si-NKILA组NKILA mRNA表达分别为1.00±0.05,1.99±0.10,2.04±0.11和1.19±0.12;细胞活力分别为(100.00±7.13)%,(54.21±5.25)%,(52.31±6.63)%和(81.05±7.89)%;细胞凋亡率分别为(4.63±0.59)%,(19.94±1.06)%,(19.67±1.76)%和(10.01±0.96)%。Control组与GSTT组比较,GSTT+si-NC与GSTT+si-NKILA组比较,差异均有统计学意义(均P<0.05)。结论蒺藜皂苷通过调控lncRNA NKILA,能够抑制膀胱癌细胞增殖,促进细胞凋亡。 Objective To investigate the effect of Tribulus terrestris saponin on the proliferation and apoptosis of bladder cancer cells and its mechanism.Methods Bladder cancer cells J82 were divided into control group,GSTT group,GSTT+si-NC group and GSTT+si-NKILA group.Control group was treated with 0 mg·L^(-1) Tribulus tribulus saponin,GSTT group was treated with 80 mg·L^(-1) Tribulus tribulus saponin.siRNA control and NKILA siRNA were transfected into the cells,and the cells were cultured with culture medium containing 80 mg·L^(-1) Tribulus tribulus saponin,as GSTT+si-NC group and GSTT+si-NKILA group.Cell viability of J82 cells was detected by cell counting kit-8(CCK-8).The expression level of NKILA was detected by real-time fluorescence quantitative reverse transcription polymerase chain reaction(RT-qPCR).The proliferation ability of each group was detected by clonal formation assay.The apoptosis of J82 cells was detected by flow cytometry.Western blot was used to detect apoptosis protein levels.Results NKILA mRNA expression in control group,GSTT group,GSTT+si-NC group and GSTT+si-NKILA group were 1.00±0.05,1.99±0.10,2.04±0.11 and 1.19±0.12;cell viability were(100.00±7.13)%,(54.21±5.25)%,(52.31±6.63)%and(81.05±7.89)%;apoptosis rates were(4.63±0.59)%,(19.94±1.06)%,(19.67±1.76)%and(10.01±0.96)%.Compared with control group and GSTT group,compared with GSTT+si-NC group and GSTT+si-NKILA group,the differences were all statistically significant(all P<0.05).Conclusion Tribulus terrestris saponins can inhibit the proliferation and promote the apoptosis of bladder cancer cells by regulating lncRNA NKILA.
