摘要
目的基于GC-TOF-MS的代谢组学技术,探索结香花不同花期差异性代谢产物及变化。方法利用GC-TOF-MS的代谢组学技术结合多元统计分析,筛选结香花不同花期差异性代谢产物及其代谢通路。结果基于正交偏最小二乘判别法分析,共鉴定出62个差异性代谢产物(VIP>1,P<0.05),进一步结合差异倍数(FC>2或<0.5)筛选出显著性差异代谢产物12个,分别为:柠檬酸、亚油酸、赖氨酸、2-脱氧叶黄素、蛋氨酸、果糖、麦芽糖、异麦芽糖、异麦芽酮糖醇、苯甲醇、棕榈酸单甘油酯、儿茶素。富集通路有乙醛酸和二羧酸代谢,赖氨酸生物合成,甘氨酸、丝氨酸和苏氨酸代谢等通路。结论基于GC-TOF-MS的代谢组学方法适用于结香花不同花期差异性代谢产物筛选,为确定结香花药材最佳采收期和质量评价提供参考和借鉴。
Objective To explore differential metabolomics for identifying flower of Edgeworthia chrysantha Lindl.from different flowering periods by gas chromatography time of flight mass spectrometry(GC-TOF-MS).Methods The metabolite profile is determined using GC-TOF-MS,then combined with multivariate statistical analysis to characterize different metabolomics between different flowering periods and explore metabolic pathway.Results A total of 62 metabolites are identified as differential metabolites(VIP>1,P<0.05)based on orthogonal partial least squares discriminant analysis,further,12 significant differential metabolites are selected by using fold change(FC>2 or<0.5),including citric acid,linoleic acid,fructose,2-deoxyerythritol,maltose,lysine,isomaltose,methionine,palatinitol,benzyl alcohol,monopalmitin,(+)-catechin.Pathway analysis of differentially metabolites show that glyoxylate and dicarboxylate metabolism;valine,leucine and isoleucine biosynthesis;lysine biosynthesis;citrate cycle(TCA cycle);starch and sucrose metabolism;glycine,serine and threonine metabolism.Conclusion Metabolites methodology based on GC-TOF-MS can be used to screening the differential metabolites of flower of Edgeworthia chrysantha Lindl.at different flowering periods,which provide a reference for harvest time and quality evaluation for medicinal herb.
