黄芪总皂苷通过激活Nrf2/HO-1信号通路缓解L-精氨酸诱导的小鼠急性胰腺炎氧化损伤的研究 被引量:2

Study on the Effect of Astragalus Saponins on Oxidative Damage of L-arginine Induced Acute Pancreatitis in Mice by Activating Nrf2/HO-1 Signaling Pathway

摘要 目的探讨黄芪总皂苷对急性胰腺炎小鼠氧化损伤的影响及其可能作用机制。方法选取65只小鼠腹腔注射L-精氨酸建立急性胰腺炎(AP)模型,造模成功小鼠随机分为模型组、黄芪总皂苷低、中、高剂量组(25、50、100 mg/kg)各15只,另设对照组15只。检测小鼠AMY、氧化因子、胰腺病理变化、Nrf2、HO-1 mRNA和蛋白表达情况等。结果与模型组比,黄芪总皂苷低、中、高剂量组胰腺指数、AMY、MDA水平降低(P<0.05),GSH-Px、SOD、CAT、Nrf2和HO-1 mRNA和蛋白表达水平升高(P<0.05),以黄芪总皂苷高剂量组效果最佳。结论黄芪总皂苷可降低AP小鼠AMY水平,改善氧化损伤,可能与激活Nrf2/HO-1信号通路有关。 Objective To investigate the effect of Astragalus saponins on oxidative damage in mice with acute pancreatitis and its possible mechanism.Methods 65 mice are injected intraperitoneally with L-arginine to establish acute pancreatitis(AP)model.The successful mice are randomly divided into model group,low,medium and high dose of Astragalus saponins groups(25,50,100 mg/kg)with 15mice in each group,another control group of 15 is set up.The levels of AMY,histopathological changes of pancreas,oxide factor,Nrf2 and HO-1 mRNA and protein expression are detected.Results Compared with the model group,the levels of pancreatic index,serum AMY and MDA are decreased(P<0.05),the levels of GSH-Px,SOD,CAT,Nrf2,HO-1 mRNA and protein expression are increased(P<0.05)in the low,medium and high dose of astragalus saponins groups.The effect of high dose of astragalus saponins group is the best.Conclusion Astragalus saponins can reduce the level of AMY in AP mice and improve oxidative damage to a certain extent,which may be related to the activation of Nrf2/HO-1 signal pathway.
