摘要
目的建立益胃汤HPLC指纹图谱,比较益胃汤标准汤剂与配方颗粒的差异,为益胃汤的质量评价提供思路。方法制备10批益胃汤标准汤剂、10批益胃汤配方颗粒剂、4批单味药材汤剂、3批益胃汤共煎配方颗粒剂,采用HPLC法建立益胃汤指纹图谱,通过相似度、毛蕊花糖苷含量、多元统计分析比较益胃汤标准汤剂与配方颗粒的差异。结果标定了20个特征峰,指认15号峰为毛蕊花糖苷。10批标准汤剂的共有模式与10批配方颗粒汤剂及3批益胃汤共煎配方颗粒剂的相似度为0.962~0.996。与配方颗粒相比,标准汤剂缺少2、4号峰,部分样品缺少5、7、9号峰。标准汤剂中毛蕊花糖苷含量高于配方颗粒。聚类分析(CA)、主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)均能区分二者。结论指纹图谱与化学模式识别结合可全面评价益胃汤标准汤剂与配方颗粒差异,为益胃汤的临床合理应用提供科学依据。
Objective To establish the HPLC fingerprint of Yiweitang,compare the differences between Yiweitang standard decoction and formula granules,and provide a method for the quality evaluation of Yiweitang.Methods 10 batches of Yiweitang standard decoction,10 batches of Yiweitang formula granules,4 batches of single herb decoction and 3 batches of Yiweitang co-decocted formula granules are prepared.The fingerprint of Yiweitang is established by HPLC.The differences between Yiweitang standard decoction and formula granules are compared by similarity,verbascoside content and multivariate statistical analysis.Results 20 characteristic peaks are calibrated and No.15 peak is identified as verbascoside.The similarity among the common mode of standard decoction,10 batches of formula granule decoction and 3 batches of co-decocted formula granule is 0.962~0.996.Compared with formula granule group,the standard decoction group lacked peaks 2 and 4,some samples lacked peaks 5,7 and 9.The average concentration of verbascoside in the standard decoction group is higher than that in the formula granules.Cluster analysis(CA),principal component analysis(PCA),orthogonal partial least squares discriminant analysis(OPLS-DA)can clearly distinguish the two groups.Conclusion The combination of chemical pattern recognition and fingerprint can comprehensively evaluate the difference between Yiweitang standard decoction and formula granules,which can provide scientific basis for clinical rational citation of Yiweitang.
