基于STR分型技术的黄牛亲权鉴定复合扩增体系的建立

摘要 目的建立用于黄牛STR检测的23个位点复合扩增体系,并对其进行性能评估与实际样本测试。方法采用荧光标记技术,选择23个黄牛基因座,进行引物设计、复合扩增与电泳检测。验证该系统的特异性、灵敏度及耐受性,并在实际样本测试中验证基因座的多态性。结果采用本系统,分型结果特异性好、灵敏度高、耐受性强,在实际样本各基因座多态性好。结论本研究建立的复合扩增体系分型结果良好,可满足实验室日常对黄牛亲权鉴定的需求。 Objective To establish and validate a novel multiplex PCR system comprised of 23 loci in cattle.Methods 23 loci used to detect cattle was amplified simultaneously in a multiplex PCR system using the labeling fluorescence.PCR product was analyzed in a ABI 3500XL Genetic Analyzer.The specifity,sensitivity and stability of the system and its application to actual samples were evaluated.Results The results demonstrated that the multiplex PCR system was stable,accurate,and sensitive.The polymorphism of each loci in the actual sample was good.Conclusion The multiplex PCR system established in this study is effective and reliable,which can meet the daily requirements of the laboratory for the paternity identification of cattle.
