摘要
为研究核桃青皮对急性高脂血症小鼠的降脂作用。实验小鼠根据体重随机分为6组:正常组、模型组、非诺贝特组和核桃青皮提取物低、中、高剂量组。各组小鼠连续给药7 d后,模型组和核桃青皮提取物组每只小鼠腹腔注射0.5 mL的75%蛋黄乳,正常组小鼠给予等量的生理盐水。小鼠在给予蛋黄乳后禁食不禁水16 h,通过摘眼球取血,用试剂盒测定血清中总胆固醇、甘油三酯、低密度脂蛋白胆固醇和高密度脂蛋白胆固醇水平。并用高效液相色谱-质谱法鉴定核桃青皮提取物中的降脂成分。实验显示非诺贝特和核桃青皮提取物能显著降低小鼠血清中总胆固醇、甘油三酯、低密度脂蛋白胆固醇水平(P<0.01),显著升高高密度脂蛋白胆固醇水平(P<0.01)。通过高效液相色谱-质谱分析,从核桃青皮提取物中鉴定出16种多酚类物质。结果表明核桃青皮提取物具有良好的降脂作用,且降脂物质可能是其中的多酚类物质。
To study the lipid-lowering effect of walnut green husk on mice with acute hyperlipidemia.The experimental mice were randomly divided into the following groups according to body weight:normal,model,fenofibrate groups,and walnut green husk extract low-dose,medium-dose,high-dose groups.After seven days of continuous administration,each mouse in the model group and walnut green husk extract group was intraperitoneally injected with 0.5 mL 75% yolk milk,and mice in the normal group were injected with the same amount of saline.The mice were then fasted without water for 16 h,after which blood was extracted from the eyeballs.The serum levels of total cholesterol,triglyceride,and low-and high-density lipoprotein cholesterols were determined using the kit.High-performance liquid chromatography-mass spectrometry was used to identify the lipid-lowering components of walnut green husk extract.The results showed that fenofibrate and walnut green husk extract significantly reduced the serum levels of total cholesterol,triglyceride,and low-density lipoprotein cholesterol(P<0.01)and significantly increased the serum level of high-density lipoprotein cholesterol(P<0.01).Sixteen polyphenols were identified from walnut green husk extract by high-performance liquid chromatography-mass spectrometry.The lipid-lowering effect observed in this study may be caused by polyphenols in the walnut green husk extract.
